Description
Die Erfindung betrifft quervernetzte hydrophile Polymere, die geeignet sind für die Verwendung als Träger für Arzneimittel
und andere aktive Bestandteile; als hydrophile Membranen für Trennverfahren; als Verbandmaterial für Wundbehandlungen; als
Körperimplantationsmaterialien; z. B. künstliche Venen und Überzüge auf Glas, Metall, Holz oder Keramikmaterialien sowie
Überzüge auf Samen und insbesondere für die Verwendung bei Anwendungsgebieten, bei denen gleichzeitig eine Festigkeit
des polymeren Gegenstands und eine hohe Permeabilität gegenüber Wasser und Sauerstoff erforderlich sind, wie bei Kontaktlinsen.
Hydrogele wurden seit 1956 beschrieben (US-PS 2 976 576) und
anschließend erschien eine große Anzahl an Patenten, die die Synthese und die Verwendung von Hydrogelen in erster Linie
auf Basis von 2-Hydroxyäthylmethacrylat und in einem geringeren
Ausmaß von H-Vinylpyrrolidon beschreiben. Typischerweise
sind diese Hydrogele quervernetzte, mit Wasser quellbare Polymere, die durch Copolymerisation von 2-Hydroxyäthylmethacrylat
hergestellt werden. Sie werden als polymere inerte Träger für aktive Substanzen verwendet, die langsam und kontrollierbar
aus diesen Trägern freigegeben werden; derartige aktive Substanzen können Wirkstoffe bzw. Arzneimittel
(US-Patentschriften 3 577 826, 3 577 512, 3 551 556, 3 520 949,
3 576 760, 3 641 237, 3 660 563), landwirtschaftliche Chemikalien (US-PS 3 576 260) oder Duftstoffe (US-Patentschriften

3 400 890, 3 567 118, 3 697 643) sein.
Ihre Verwendung als ein Beschlagen verhindernde Überzüge (US-PS 3 488 215), Körperimplantationsmittel und Verbandmaterial
wurde auch in den US-Patentschriften 3 577 516, 3 695 921, 3 512 183, 3 674 901 beschrieben. Die in weitem
Umfang verwendeten weichen Kontaktlinsen bestehen aus diesem Material (US-Patentschriften 3 488 111, 3 660 545,
3 629 504).
Auf dem pharmazeutischen Gebiet liegt das Hauptinteresse bei der langsamen und kontrollierbaren !freigäbe von Wirkstoffen
aus derartigen Hydrogelen. Als Wirkstoff enthaltende Hydrogelpreparate
wurden solche in Form von Verbandmaterialien, subkutanen Implantaten, buccalen Vorrichtungen, Intrauterinvorrichtungen
und Augeneinlagen beschrieben. Sie werden nach komplizierten Herstellungsverfahren hergestellt, die gewöhnlich
das Gießen der Monomerenlösung in eine geeignete
Form und die Polymerisation in Anwesenheit eines freien Radikale bildenden Initiators umfassen.
Die Verwendung von wirkstoffbeladenen Hydrogelgranulaten als
orale Dosisform wurde auch in der GB-PS 1 135 966 und der US-PS 3 551 556 empfohlen. Sie ist in der Tat eine der
nützlichsten Anwendungen auf diesem Gebiet in der Medizin, da sie die Abgabe eines oral eingenommenen Wirkstoffs an den
Blutstrom unter Ausbreitung während mehrerer Stunden in reproduzierbarer Weise ermöglicht. Hierdurch werden unnötige
und möglicherweise gefährliche Wirkstoffspitzenkonzentrationen in dem Blut vermieden, wobei die Dauer,während der bevorzugte
und wirksame Wirkstoffspiegel in dem Blut aufrechterhalten werden, verlängert wird.
Es sind mäßig quervernetzte wasserunlösliche hydrophile Polymere bekannt, die durch Copolymerisation einer größeren Menge
eines hydrophilen monoolefinischen Monomeren und einer geringeren Menge im Bereich von 0,01 bis 15 i° dieses Monoolefins

eines Quervernetzungsmittels mit niedrigem Molekulargewicht hergestellt werden.
Diese hydrophilen monoolefinischen Monomere sind im allgemeinen die Monoester der* Acryl- oder Methacrylsäure mit polyfunktionellen
Alkoholen. 2-Hydroxyäthylmethacrylat wird in besonders großem Umfang verwendet. Die Quervernetzungsmittel
sind im allgemeinen die Diester der gleichen Säuren mit den gleichen polyfunktionellen Alkoholen und Äthylenbisme-thacrylat
wird in besonders großem Umfang verwendet.
Die Copolymerisation wird in Anwesenheit von Wasser (US-PS 3 220 960) oder in einem wasserfreien System
(US-PS 3 220 949) durchgeführt. Wasserlösliche Substanzen mit
niedrigem Molekulargewicht sowie makromolekulare Substanzen, wie Poly(äthylenoxyd)monomethacrylat zusammen mit
einer geringeren Menge des entsprechenden Bis-methacrylats,
wurden als Monomere und Quervernetzungsmittel verwendet (US-PS 3 220 960). Die wasserlöslichen, jedoch hydrophilen
Copolymeren und das Verfahren zu deren Herstellung wurden in verschiedenen Richtungen modifiziert und speziellen Zwecken
z. B. der Herstellung von weichen Kontaktlinsen (US-PS 3 200 960 und Reissue Kr. 27 4-01) und der Copolymerisation
in Anwesenheit eines linearen Polyamidharzes zur Verbesserung oder Modifizierung der mechanischen Eigenschaften
von Formkörpern, die aus den erhaltenen Polymeren geformt wurden (US-PS 3 520 949), angepasst. Jedoch wurden bei
sämtlichen Modifikationen polyolefinische Quervernetzungsmittel mit niedrigem Molekulargewicht, insbesondere Äthylenbismethacrylat,
in sehr geringen bis mäßigen Mengen, die niemals 20 $ der Menge des monoolefinischen Monomeren überschritten,
verwendet. Obgleich die Copolymeren des vorstehend beschriebenen Typs modifiziert werden könnten, um den
Anforderungen von mehreren verschiedenen Verwendungsmethoden derselben zu entsprechen, konnten die mechanischen Eigenschaften
im entweder ungequollenen, d. h. wasserfreien, oder

gequollenen Zustand, ά. Ii. in* Gleichgewichtszustand mit Wasser,
nicht zufriedenstellend angepasst werden, um sämtlichen Anforderungen
für die endgültige Verwendung zu entsprechen. Es ist bekannt, daß hydrophile Polymere, deren überwiegende
Bestandteile Monoester der Acrylsäure und der Methacrylsäure
und eines bifunktionellen Alkohols (Glykol) sind, Glasübergangstemperaturen
oder Erweichungspunkte zwischen 550C und 800C besitzen. Aus diesem Grunde sind die Gegenstände des
Stands der Technik in trockenem Zustand bei Temperaturen unterhalb 55°C brüchig und glasig. Nach der Gleichgewichtseinstellung in Wasser werden diese Gegenstände aus dem Stand
der Technik weich und etwas biegsam, jedoch auch im Hinblick
auf ihre Biegeeigenschaften schwach. Zusätzlich sind diese Gegenstände aus dem Stand der Technik im hohen Ausmaß
reißenden Scherkräften zugänglich, wenn sie auf irgendeine Weise versehentlich beschädigt werden. Insbesondere sind sie
in gequollenem Zustand außerordentlich brüchig.
Um die unerwünschten schwachen Eigenschaften der aus diesen Polymeren des Stands der Technik hergestellten Gegenstände
zu vermeiden, wird ein aus stärkerem Polymerenmaterial hergestelltes Medium als physikalischer Träger verwendet, oder
die prepolymerisierte Mischung wird mit einem unlöslichen Material, wie Silicagel angereichert. Diese Techniken ergeben,
obgleich sie zu einem bestimmten Ausmaß eine kohäsive Festigkeit verleihen (wobei das Hydrogelmaterial als Klebmittel
wirkt), Gegenstände, die noch in trockenem Zustand einem glasartigen Bruch und einem Scherbruch in gequollenen Zustaid innerhalb
der Zwischenraumbereiche des Gegenstands zugängig sind.
Aus dem gleichen Grund modifiziert die Zugabe von Füllstoffen
zu dem Prepolymeren die Diffusionseigenschaften und die Wasserpermeabilität des Gegenstands häufig in unerwünschter
Weise. Natürlich kann ein opakes Polymeres nicht als Kontaktlinsenmaterial
verwendet werden.
Die DE-OS 2 528 068 beschreibt eine neue Klasse von Hydrogelmaterialien,
die diese Nachteile nicht aufweisen,die jedoch

in trockenem sowie in gequollenem Zustand zäh und flexibel sind. Dies wird durch Einarbeiten einer größeren Menge eines
hydrophilen Quervernetzungsmittels mit hohem Molekulargewicht erreicht, was eher makromere, flexible Quervernetzungen als
übliche kurze Quervernetzungen, die zu Brüchigkeit führen, ergibt. Für viele Anwendungen war noch die Säuerstoffpermeabilität
zu niedrig, insbesondere für Zusammensetzungen mit einem niedrigen Gleichgewichtswassergehalt. Die Kombination
eines niedrigen, jedoch merklichen Wassergehalts, und einer hohen Säuerstoffpermeabilität ist besonders auf dem Gebiet
der weichen Kontaktlinsen wichtig. Kontaktlinsen sollten einen Wassergehalt aufweisen, der ausreichend hoch ist, um
ihr Tragen angenehm zu machen, der jedoch nicht zu hoch ist, um zu optischen Verzeichnungen zu führen. Andererseits sollten
sie eine ausreichend hohe Säuerstoffpermeabilität bei derart
niedrigen Quellungsgraden aufweisen. Gleichzeitig sollten sie zäh und flexibel sein und auch noch geeignet für eine
maschinelle Bearbeitung und Politur.
Es war seit langem bekannt, daß Polymere auf Siliconbasis eine hohe Säuerstoffpermeabilität besitzen und Kontaktlinsen
auf Siliconbasis wurden in der GB-PS 1 170 810 und der PR-PS 1 526 934 sowie in der DE-OS 2 228 180 beschrieben.
In all diesen Fällen bildet das Siliconpolymere den Kern einer Kontaktlinse, deren Oberfläche speziell behandelt wurde, um
sie hydrophil und mit der Hornhaut des Auges verträglich zu machen. Dies wird durch Aufpfropfen eines hydrophilen Monomeren
auf die Kontaktlinsen unter Verwendung einer freie Radikale erzeugenden Bestrahlung in Anwesenheit oder Abwesenheit
von Sauerstoff erreicht. Dies kann entweder während oder nach der Herstellung der Kontaktlinsen erfolgen, jedoch
handelt es sich in beiden Fällen um ein kompliziertes und schwierig kontrollierbares Verfahren. Es muß die Diffusion
des Monomeren in den Kern verhindert werden, da sie zu einer Trübung führt und jede Linse getrennt geformt werden muß, und
da der weiche und zerbrechliche Polysiloxankern nicht maschinell bearbeitet werden kann. Die Möglichkeit einer

maschinellen Bearbeitung und einer Politur einer Eontaktlinse
ausgehend von einem kleinen unbearbeiteten Werkstück stellt einen großen Vorteil dar, da sie es ermöglicht, die Linsen den
individuellen Anforderungen mit großer Präzision anzupassen.
Es wurde nun gefunden, daß eine unerwartet ideale Kombination
von wertvollen Eigenschaften erreicht werden kann, indem man ein Hydrogel synthetisiert, das einen größeren Anteil eines
Quervernetzungsmittels auf Basis eines polymeren Polysiloxans
enthält. Diese Eigenschaftenkombination umfaßt: Hydrophilie, Säuerstoffpermeabilität, Flexibilität und Zähigkeit in
trockenem und gequollenem Zustand, Bioverträglichkeit, Polierbarkeit und mechanische Bearbeitbarkeit im allgemeinen,
Klarheit und eine einfache und wirtschaftliche Synthese.
Ziel der Erfindung ist die Schaffung von quervernetzten, hydrophilen Copolymeren mit verbesserter Sauerstoffpermeabilität
und Gewebeverträglichkeit, die eine hohe Flexibilität und hohe Elastizität sowohl in im wesentlichen wasserfreien
Zustand als auch in gequollenem Zustand, d. h. im Gleichgewicht mit Wasser oder wässrigen Flüssigkeiten, wie Körperflüssigkeiten
von Lebewesen^aufweisen, die geeignet sind für die Verwendung als hydrophile Membranen für Gas- und
Flüssigkeitstrennverfahren, Verbandmaterial zur Wundbehandlung; Körperimplantate, z. B. künstliche Venen, Kontaktlinsen;
Überzüge auf Glas, Metall, Holz oder Keramik sowie Überzüge auf Samen und als Träger für biologisch aktive Substanzen,
z. B. Arzneimittel, Herbizide, Insektizide, Fungizide, Bakterizide und Duftstoffe.
Ein weiteres Ziel der Erfindung ist die Schaffung von quervernetzten,
hydrophilen Copolymeren, die die vorstehende Eignung besitzen und eine hohe Zugfestigkeit in gequollenem
Zustand aufweisen.
Ein anderes Ziel der Erfindung ist die Schaffung von quervernetzten,
hydrophilen Copolymeren, die die vorstehend ange-

gebenen verbesserten Eigenschaften besitzen, und eine variierbare wirksame Menge an biologisch aktiven Materialien,
insbesondere therapeutisch aktiven Substanzen, enthalten.
Es wurde nun gefunden, daß wasserunlösliche hydrophile Copolymere,
die im wesentlichen aus einem hydrophilen Polymeren von monoolefinischen Monomeren, die mit einer größeren Menge
eines Makromeren auf Basis eines di- oder triolefinischen Polysiloxans quervernetzt sind, bestehen und eine biologisch
aktive Substanz enthalten können, die vorstehend angegebenen und weitere wünschenswerte Eigenschaften besitzen. Bei den
neuen Copolymeren ist der Anteil des endständigen di- oder triolefinischen Siloxanmakromeren ein größerer Anteil des
Systems der bis zu ca. 10 bis ca. 80 fa des hydrophilen Polymeren
oder vorzugsweise von ca. 15 bis 60 $ beträgt. Dieses
endständige di- oder triolefinische Siloxanmakromere besitzt
ein Molekulargewicht zwischen ca. 400 und ca. 8500 oder vorzugsweise zwischen ca. 600 und ca. 5000.
Die Erfindung betrifft ein wasserunlösliches hydrophiles Gel, das das quervernetzte Copolymerisationsprodukt von (A) ca.
20 bis ca. 90 Gew.-$ dieses Gels eines (a) wasserlöslichen
monoolefinischen Monomeren oder einer Mischung dieser Monomeren oder (b) eines wasserlöslichen Monomeren oäer einer
Mischung dieser Monomeren mit 1 bis 80 Gew.-^ der Gesamtmonomeren
eines wasserunlöslichen monoolefinischen Monomeren oder einer Mischung dieser wasserunlöslichen Monomeren mit
(B) ca. 10 bis ca. 80 Gew.-$ dieses Gels eines endständigen
polyolefinischen hydrophoben Siloxanmakromeren mit einem Molekulargewicht von ca. 400 bis ca. 8500 umfaßt, wobei dieses

Die neuen erfindungsgemäßen Hydrogele werden hergestellt durch
Copolymerisation mit Hilfe von freien Radikalen, entweder in Lösung oder in der Masse eines wasserlöslichen Monomeren
(As), das ein wasserlösliches oder in Wasser quellbares Polymeres bilden kann, d. h. ein hydrophiles Polymeres (Ap), mit
einem sich von einem di- oder triolefinischen Siloxan ableitenden Makromeren (B), z. B. einer "Verbindung, bestehend aus
einer langen linearen Polysiloxankette, die an beiden Enden mit einer polymerisierbaren Yinylgruppe endständig versehen
ist. Auf diese Weise wird ein dreidimensionales makromolekulares Netzwerk gebildet, das aus zwei Segmenttypen
besteht, wobei jedes Segment dem System seine bestimmten physikalischen Eigenschaften verleiht. Das Α-Segment verleiht
Wasserlöslichkeit; das Siloxan-B-Segment bildet flexible
Quervernetzungen und verbessert die Säuerstoffpermeabilität
und die Gewebeverträglichkeit. Indem man die relativen Anteile
einer jeden Verbindung variiert, kann ein weiter Bereich an mechanischen und Diffusionseigenschaften erhalten
werden. Beispielsweise können Polymere mit ausgezeichneter Festigkeit und Dehnung sowie Zähigkeit, die auch noch so viel .
wie annähernd ihr eigenes Gewicht an Wasser absorbieren können, erhalten werden, indem man die geeigneten relativen Anteile
von A und B verwendet. Dies steht im Gegensatz zu herkömmlichen Hydrogelen, deren Quervernetzungen kurz und nicht
elastisch sind, und die in trockenem Zustand hart und brüchig sind.
Die Neuheit der erfindungsgemäßen Polymeren beruht auf der Einarbeitung des di- oder trifunktionellen Makromoleküls B
als größeren Anteil dieses Systems. Auf diese Weise dient das difunktionelle Makromere nicht nur als strukturelle Quervernetzung,
sondern verleiht auch dem Gel seine einzigartigen

physikalischen Eigenschaften.
Das hydrophile Polymere Ap ist vorzugsweise ein Polymeres von einem oder mehreren wasserlöslichen monoolefinischen Monomeren
As, jedoch kann es auch ein Copolymeres von monoolefinischen wasserlöslichen Monomeren mit höchstens 30 <fo, vorzugsweise
höchstens 40 $, der Gesamtmenge der Monomeren eines wasserunlöslichen monoolefinischen Monomeren Ai sein.
Die wasserlöslichen Monomeren As sind vorzugsweise Aeryl-
und/oder Methacrylsäure oder deren wasserlösliche Derivate, wie die Hydroxyalkylester, worin Alkyl 2 bis 4 Kohlenstoffatome
aufweist, z. B. 2-Hydroxyäthyl-, 3-Hydroxypropyl-, 2-Hydroxypropyl- oder 2,3-Dihydroxypropylester; sowie äthoxylierte
und polyäthoxylierte Hydroxyalkylester, wie Ester von Alkoholen der Formel
worin m 2 Ms 5 "bedeutet und η 1 Ms 20 bedeutet
oder Ester von analogen Alkoholen, worin ein Teil der Äthylenoxydeinheiten
durch Propylenoxydeinheiten ersetzt ist. Ebenfalls geeignet sind 3-(Dimethylamino)-2-hydroxypropy!ester
und -amide. Eine weitere Klasse an geeigneten Derivaten derartiger Säuren sind ihre wasserlöslichen Amide, wie die unsubstituierten
Amide und die durch Hiedrighydroxyalkyl-,
Niedrigoxaalkyl- oder Niedrigdialkylaminoalkylgruppen substituierten
Amide, worin Alkyl 2 bis 4 Kohlenstoffatome aufweist, wie F-(Hydroxymethyl)-acrylamid und -methacrylamid,
N-(3-Hydroxypropyl)-acrylamid, N-(2-Hydroxyäthyl)-methacrylamid
und N-[1,1-Dimethyl-2-(hydroxymethyl)-3-oxabutyl!-acrylamid;
wasserlösliche Hydrazinderivate, wie Dimethyl-2-hydroxypropylaminmethacrylimid
und die entsprechenden Derivate der Acrylsäure.
Ebenfalls in Kombination mit Comonomeren verwendbar sind die

Hydroxyalkylmonomaleate, wie 2-Hydroxyäthylmonomaleat und alkoxylierte Hydroxyalkylmonomaleate
niit Vinyläthern, VinyIestern, Styrol oder allgemein
jedem Monomeren, das leicht mit Maleaten oder Fumaraten
copolymerisiert; Hydroxyalkylvinylather, wie 2-Hydroxyäthylvinylather,
4-Hydroxybutylvinylather mit Maleaten, Fumaraten
oder allgemein mit allen Monomeren, die leicht mit Vinyläthern copolymerisieren.
Besonders wertvoll als wasserlösliche Monomere sind die Hydroxyalkylacrylate und -methacrylate, wie 2-Hydroxyäthy1-acrylat,
2-Hydroxyäthylmethacrylat, 2-Hydroxypropylacrylat
3-Hydroxypropylmethacrylat, 2,3-Dihydroxypropylmethacrylat.
Bevorzugte hydroxysubstituierte Viny!monomere sind 2-Hydroxyäthylmethacrylat
und 2- und 3-Hydroxypropylmethacrylat.
Am bevorzugtesten ist 2-Hydroxyäthylmethacrylat.
Wasserlösliche Comonomere, die keine Hydroxygruppen enthalten,
sind: Acryl- und Methacrylsäure und. Alky lather von polyäthoxylierten
Hydroxyalkylestern derselben, wie die Ester von Alkoholen der Formel
worin m 2 bis 5 bedeutet und η 4 bis 20 bedeutet.
Dialkylaminoalkylester und -amide, wie 2-(Dimethylamine)-äthyl-
oder 2-(Diathylamino)-äthylacrylat und -methacrylat sowie die
entsprechenden Amide; Amide substituiert durch Niedrigoxaalkyl-
oder Niedrigdialkylaminoalkylgruppen, wie N-(1,1-Dimethy1-3-oxa-butyl)-acrylamid;
wasserlösliche Hydrazinderivate, wie Trialkylaminraethacrylimid, z. B. Triäthylaminmeth-

Monoolefinische Sulfonsäuren und deren Salze, wie Natriumäthylensulf

heterocyclischen Stickstoff enthaltenden Monomeren, wie H-Vinylpyrrol, N-Vinylsuccinimid, i-Vinyl-2-pyrrolidon,
1-Vinylimiäazol, 1-Yinylindol, 2-Vinylimidazol,
4(5)-Vinylimidazol, 2-Vinyl-i-methylimidazol, 5-Vinylpyrazolin,
3-Methyl-5-isopropenylpyrazol, 5-Methylenhydantoin,
3-Vinyl-2-oxazolidon, 3-Methacrylyl-2-oxazolidon, 3-Methacrylyl-5-methyl-2-oxazolidon, 3-Vinyl-5-methy1-2-oxazolidon,
2- und 4-Vinylpyridin, 5-Vinyl-2-methylpyridin, 2-Vinylpyridin-1-oxid, 3-Isopropenylpyridin,
2- und 4-Vinylpiperidin, 2- und 4-Yinylchinolin, 2,4-Dimethyl-6-vinyl-s-triazin
und 4-Acrylylmorpholin.
Unter diesen Monomeren sind "bevorzugt N-Vinyl-2-pyrrolidon,
2-Vinylpyridin, 4-Yinylpyridin, 2-(Dimethylamino)-äthylmethacrylat,
Methacrylamid, Acrylsäure und Methacrylsäure.
Am bevorzugtesten ist N-Vinyl-2-pyrrolidon.
Diese Monomeren können allein oder in Kombination miteinander
und anderen geeigneten Vinylmonomeren, die auch hydrophob sein können, verwendet werden. Die Menge dieser hydrophoben
Monomeren soll 72 $ der &esamtzusammensetzung nicht überschreiten
und beträgt vorzugsweise weniger als 40 $.
Geeignete hydrophobe Comonomere Ai, die in das Hydrogel eingearbeitet
werden können, sind z. B. wasserunlösliche olefinische
Monomere, wie Alkylacrylate oder -methacrylate, worin Alkyl 1 bis 18 Kohlenstoffatome aufweist, z. B. Methyl- u.
Äthylmethacrylat oder -acrylat; sich von Alkanearbonsäuren
mit 1 bis 5 Kohlenstoffatomen ableitende Vinylester, z. B. Vinylacetat und Vinylpropionat oder Vinylbenzoat; Acrylnitril;
Styrol; und Vinylalkylather, worin der Alkylteil der A'therkette 1 bis 5 Kohlenstoff atome aufweist, z. B. Methyl-,

Äthyl-, Propyl-, Butyl- oder Amylvinylather.
Bevorzugt sind Alkylacrylate und Alky!methacrylate, worin die
Alkylgruppe 1 bis 5 Kohlenstoffatome aufweist; Vinylacetat;
Styrol; und Acrylnitril.
Der hydrophile Teil der Hydrogelzusammensetzung wird durch
Polymerisation eines wasserlöslichen monoolefinischen Monomeren oder einer Mischung dieser Monomeren, die 0 bis 80
Gew.-^ der Gesamtmenge der Monomeren eines wasserunlöslichen
Monomeren enthalten kann, hergestellt.
In dem endständigen diolefinischen makromeren hydrophoben Quervernetzungsmittel (B) wurden dessen Olefinteile vorzugsweise
von Acylgruppen von niedrigen a,ß-monoungesättigten aliphatischen Monocarbonsäuren oder Dicarbonsäuren oder von
Yinyloxyteilen geliefert. Diese Vinylteile werden mit Hilfe
einer verknüpfenden Gruppe, wie sie durch ein Diisocyanat
geliefert wird, an eine sich wiederholende Siloxangruppen enthaltende makromolekulare Kette gebunden. Das Molekulargewicht
des Makromeren kann von ca. 400 bis ca. 8500, vorzugsweise zwischen ca. 600 und 8500, und insbesondere zwischen 1500 und
3000 variieren. Somit entspricht das Makromere vorzugsweise der Forme1

Verbindungen der Formel B, bei denen Y eine direkte Bindung darstellt, sind Polyester von makromolekularen PoIyolen,
worin Hydroxygruppen an die Polykondensatkette R1 gebunden
sind, mit α,β-ungesättigten Säuren. Derartige Ester können aus dem makromolekularen Polyol nach allgemein bekannten
Acylierungsmethoden unter Verwendung von reaktiven funktioneilen Derivaten von geeigneten Säuren, z. B.
Chloriden der Acryl- oder Methacrylsäure, oder Monoalkylestern der Malein-, Fumar- oder Itaconsäure; oder vom Anhydrid
der Malein- oder Itaconsäure, hergestellt werden.
Gemäß der Definition der Formel B kann Y weiterhin ein zweiwertiger
Rest -R7-Z1-COFY-R8-NH-CO-Z2- sein. Hierin ist Rr7
z. B. Methylen, Propylen, Trimethylen, Tetramethylen, Pentamethylen,
Neopentylen-(2,2-dimethyltrimethylen), 2-Hydroxytrimethylen
oder 1-(Dimethylaminomethyl)-äthylen und insbesondere Äthylen. Der zweiwertige Rest Rg leitet sich von
einem organischen Diisocyanat ab und ist ein aliphatischer Rest, wie Alkylen, z. B. Äthylen, Tetramethylen, Hexamethylen,
2,2,4-TrimethyIhexamethylen, 2,4-, 4-TrimethyIhexamethylen;
Fumaroyldiäthylen oder 1-Carboxypentamethylen; ein cycloaliphatischer
Rest, z. B. 1,4-Cyclohexylen oder 2-Methyl-1,4-cyclohexylen;
und ein aromatischer Rest, wie m-Phenylen, p-Phenylen, 2-Methyl-m-phenylen, 1,2-, 1,3-, 1,5-, 1,6-, 1,7-,
1,8-, 2,3- und 2,7-Naphthylen,4-Chlor-1,2- und 4-Chlor-1,8-naphthylen,
1-Methyl-2,4-, 1-Methy1-2,7-, 4-Methyl-1,2-,

lineares oder verzweigtes Alkylen mit 2 bis 5 Kohlenstoffatomen "bedeutet. Verbindungen mit dieser verknüpfenden
Gruppe' werden durch basenkatalysierte Addition eines Alkylenbisacrylats an ein Polydimethylsxloxandithiol in einem Molverhältnis
von 2 : 1 hergestellt.
Somit sind Verbindungen der Formel B, worin Y diese zweiwertigen
Reste bedeutet, wenn X Sauerstoff bedeutet, Bisvinylather
oder, wenn X -COO- oder CONRi- bedeutet, Bisacrylate,
Bismethacrylate, Bismaleate, Bisfumarate und Bisitaconate.
R.J leitet sich insbesondere von makromeren Polyolen oder
Dithiolen mit einem Molekulargewicht (MW) von 200 bis 8000 ab.
Verbindungen, in denen Y -R7-Z1-COlJH-R8-NHCO-Z2- bedeutet,
werden in einer Zweistufenreaktion erhalten, indem man zuerst das Polydimethylsiloxandiol, -triol oder -dithiol mit ca.
der zweifachen stöchiometrischen Menge eines aliphatischen,
cycloaliphatischen oder aromatischen Diisocyanate, das aus 2 Isöcyanat (NCO)-Gruppen, die an den Rest Rg gebunden sind,
besteht, umsetzt, um jede der -OH- oder -SH-Gruppe in dem Siloxanmolekül mit einer Endgruppe zu versehen. In einer
zweiten Stufe wird das Isöcyanat-
endverkappte Siloxan mit einer Verbindung der

Bedeutet X Sauerstoff, so ist (C) ein Vinyläther, der aktiven

Wasserstoff enthält, ζ. B. ein Hydroxyalkylvinyläther oder ein Aminoalkylvinylather; bedeutet X -COO- oder -CONRc--, so
ist (C) ein Acrylat, Methacrylat, Maleat, Fumarat, Itaconat
oder das entsprechende Amid, das aktiven Wasserstoff in der substituierten Alkylgruppe enthält. Das makromolekulare Diol
oder Diamin wird vorzugsweise in geringem Überschuss verwendet, d. h. das Terhältnis der Isocyanatgruppen zu den
Hydroxy- oder Aminogruppen sollte während der ersten Stufe der Synthese des Makromeren zumindest 1:1 betragen, wobei es
vorzugsweise zumindest 1:1,05 beträgt. Ist die Verbindung der Formel (C), die während der zweiten Stufe der Synthese des
Makromeren verwendet wird, die gleiche wie diejenige, die als hydrophiles Monomeres verwendet wird, dann kann ein großer
Überschuss dieser Verbindung verwendet werden, derart, daß die erhaltene lösung des Makromeren (B), gelöst oder dispergiert
in dem Monomeren (A),direkt für die Herstellung des endgültigen -Hydrogels verwendet werden kann.
Die bevorzugteren Siloxanmakromeren der Formel (B) besitzen ein Molekulargewicht im Bereich von ca. 600 bis ca. 8500 und
umfassen ein Polysiloxanpolyol oder -dithiol mit einem Molekulargewicht von ca. 400 bis ca. 8000, das zuerst mit
Isophorondiisocyanat oder Tolylen-2,4-diisocyanat endständig
abgeschlossen wurde, und dann mit einer Hydroxyverbindung,
ausgewählt unter den Hydroxyalkylestern, worin Alkyl 2 bis 4 Kohlenstoffatome aufweist, der Acrylsäure, Methacrylsäure,
Maleinsäure und Fumarsäure, den Hydroxyestern dieser Säuren mit einem Alkohol der Formel

2-Hydroxyäthylmethacrylat, 2-Hydroxypropylacrylat,

2-Hydroxypropylmethacrylat, 3-Hydroxypropylacrylat,
3-Hydroxypropylmethacrylat, 2,3-Dihydroxypropylacrylat oder
2,3-Dihydroxypropylmethacrylat.
Die bevorzugteste Hydroxyverbindung ist 2-Hydroxyäthylacrylat
oder 2-Hydroxyäthylmethacrylat.
Das Polysiloxanpolyol oder -dithiol wird am bevorzugtesten
endständig mit Isophorondiisocyanat umgesetzt und die Hydroxyverbindung
ist am bevorzugtesten 2-Hydroxyäthylmethacrylat.
Die Synthese des Makromeren (B) wird geeigneterweise bei einer
Temperatur im Bereich von ca. Raumtemperatur bis ca. 800C
durchgeführt. Vorzugsweise beträgt die verwendete Temperatur nicht mehr als 40 C, und am geeignetsten liegt sie im Bereich
von ca. 30 bis 400C. Die Umwandlung der Isocyanatgruppe wird
durch Infrarotspektroskopie oder durch Titration verfolgt.
Bevorzugte Diisocyanate für die Herstellung des Makromeren sind Tolylen-2,4-diisocyanat und Isophorondiisocyanat.
Eine weitere Methode zur Herstellung des Makromeren ist die Umsetzung eines hydroxylendständigen Prepolymeren, z. B. mit
Acryloylchlorid, Methacryloylchlorid oder Maleinsäureanhydrid , wobei auf diese Weise die Bildung eines Makromeren
ohne Verknüpfung von Urethanbindungen wie z. B. eines Makromeren,
worin Y eine direkte Bindung darstellt, erfolgt.
Die Polymerisation durch freie Radikale wird durch einen Initiator in Gang gesetzt, der freie Peroxy- oder Alkylradikale
in ausreichend hoher Konzentration bilden kann, um die Polymerisation der Vinylmonomeren, die bei der Synthesetemperatur
verwendet werden, zu initiieren. Diese Initiatoren sind vorzugsweise Peroxyde oder Azokatalysatoren mit einer
Halbwertszeit bei der Polymerisationstemperatur von zumindest 20 Minuten. Typische verwendbare Peroxyverbindungen umfassen:
Isopropylpercarbonat, tert.-Butylperoctoat, Benzoylperoxyd,

2,2'-Azo-bis-(2,4-dimethyl-4-methoxyvaleronitril).
Wird die Polymerisation in Wasser durchgeführt, so können
wasserlösliche Peroxyverbindungen, wie Natrium-, Kalium- oder Ammoniumpersulfat, sowie freie Radikale bildende Redoxsysterne,
wie eine Persulfat-Bisulfitkombination verwendet werden.
Andere freie Radikale bildende Mechanismen, wie Röntgenstrahlen, Elektronenstrahlen und UV-Bestrahlung, können verwendet
werden.
Die Menge des Initiators kann von 0,02 io bis 1 # bezogen auf
das Gewicht des Monomeren (A) und Makromeren (B) variiert werden, jedoch beträgt sie vorzugsweise 0,03 bis 0,3 $ bezogen
auf das Gewicht derselben.
Die neuen erfindungsgemäßen hydrophilen Hydrogelcopolymeren
werden hergestellt durch Copolymerisation mit Hilfe von freien Radikalen, entweder in Lösung oder in Masse von wasserlöslichen
monoolefinischen Monomeren As oder einer Mischung von zumindest 20 $ wasserlöslichen Monomeren As mit zumindest
80 io wasserunlöslichen monoolefinischen Monomeren Ai mit
10 bis 80 ic bezogen auf das Gesamtgewicht des Hydrogels an
Makromeren der Struktur B. Die Polymerisation wird geeigneterweise
mit einem freie Radikale liefernden Initiator bei einer
Temperatur im Bereich von ca. 40 bis ca. 1500C durchgeführt,
wobei die bevorzugte Temperatur zwischen ca. 500C und ca.
1000C liegt. Ist eine therapeutisch oder anderweitig biologisch

aktive Substanz während der Polymerisation anwesend, so kann
deren WärmeStabilität für die Polymerisationstemperatur ein
einschränkender Faktor sein.
Eine bevorzugte Methode zur Herstellung des Hydrogelgegenstands
umfaßt das Auflösen von ca. 0,02 bis ca. 1 Gew.-^ eines
geeigneten freie Radikale bildenden Initiators in der Makromeren-Monomerenlösung
mit einem Makromeren-Monomerenverhältnis, das so gewählt ist, daß die für das Hydrogel gewünschten mechanischen und Wasserabsorptionseigenschaften
entstehen und die Polymerisation der Mischung bei beispielsweise
800G während ca. 2 Stunden in einer geschlossenen Form,
um eine flache Folie des das Arzneimittel in Form einer
quasifesten Lösung enthaltenden Hydrogels zu bilden. Die Folie wird dann einem Hochvakuum bei ca. 100 C während ca.
12 Stunden ausgesetzt oder durch Quellen in lösungsmitteln, wie Alkoholen oder Wasser, extrahiert, um verbliebene Monomere und Initiatorzersetzungsprodukte zu entfernen.
Eine bevorzugte Laboratoriumsmethode zur Herstellung des
Hydrogels in Form eines Zylinders umfaßt das Füllen eines flexiblen Polyäthylenrohrs mit der bevorzugten Zusammensetzung
des Makromeren,der Monomeren und des Katalysators und die Umsetzung der Mischung während ca. 2 Stunden bei
80 C. Der fertiggestellte Gegenstand wird durch longitudinales
Aufschlitzen des Rohres und Abstreifen von dem Hydrogelgegenstand
entfernt.
Eine weitere bevorzugte Methode für die Herstellung des Hydrogels
in Form von kleinen Kugeln oder Perlen umfaßt die Polymerisation der bevorzugten Zusammensetzung des Makromeren und
der Monomeren und des Katalysators in einem Medium, das für irgendeinen Teil der Hydrogelzusammensetzung bei ca. 30 bis
900C kein Lösungsmittel darstellt. Beispiele für geeignete
Perlpolymerisationsmedien sind Siliconöle, polyfluorierte
Öle und dergleichen, z. B. Lackbenzin bzw. Ligroin und gesättigte wässrige Salzlösungen. Im letztgenannten Fall ver-

hindern Dispergiermittel wie Poly-(viny!pyrrolidon), Hydroxyäthylcellulose,
Magnesiumhydroxyd oder Calciumphosphate eine Koagulation.
Eine weitere bevorzugte Methode zur Herstellung des Hydrogels in Form eines geschäumten Gegenstandes umfaßt das Beimischen
eines üblichen Schäumungsmittels bzw. Blähmittels, wie
Matriumbicarbonat, zu der bevorzugten Zusammensetzung und
die Polymerisation der Mischung in einer Porm bei ca. 80 C während ca. 1 Stunde. Der erhaltene gesehlossenzellige Schaum
ist besonders für eine schnelle Wasserabsorption und Arzneimittelfreigabe geeignet.
Die Reaktion wird vorzugsweise unter inerter oder anaerober Atmosphäre, sofern sie in offenen Formen erfolgt, durchgeführt.
Es ist bekannt, daß Sauerstoff die Polymerisation inhibiert und Veranlassung zu ausgedehnteren Polymerisationszeiten für eine Beendigung der Reaktion gibt. Werden geschlossene
Formen für die Formung des Hydrogelgegenstandes verwendet, so bestehen die Formen aus inerten Materialien mit
geringer Sauerstoffpermeabilität und nichtklebenden Eigenschäften.
Beispiele für geeignete Formmaterialien sind Teflon6^
Siliconkautschuk, Polyäthylen und Mylar*^. G-las- und Metallformen
können verwendet werden, wenn ein geeignetes die Form freigebendes Mittel verwendet wird.
Das Einarbeiten eines Arzneimittels in den Hvdrogelgegenstand kann entweder durch Auflösen oder Dispersion in der Makromerenlösung,
der Monomerenlösung oder einer Mischung derselben vor der Zugabe des freie Radikale liefernden Katalysators oder
durch Diffusion des Arzneimittels in den fertiggestellten Gegenstand nach der Polymerisation erfordern. Ist das Arzneimittel
gegenüber einem Angriff durch freie Radikale inert, so ist es bevorzugt, es in der Makromerenlösung, der Monomerenlösung
oder deren Mischung vor der Polymerisation aufzulösen oder zu dJMr,fi.'gieeen. Ist das Arzneimittel einem Angriff durch
freie Radikale zugänglich, so wird es in den fertiggestellten

Gegenstand nach der Polymerisation durch Diffusion aus einem Lösungsmittelmedium eingebracht.
Das im wesentlichen aus dem Siloxanmakromerensegment B und
dem hydrophilen Segment Ap bestehende Hydrogelsystem kann hinsichtlich der Zusammensetzung innerhalb eines weiten Bereichs
variieren, und demzufolge können der Grad der Hydrophilie und der mechanischen Festigkeit derart eingestellt
werden, daß zahlreichen Anwendungen Folge geleistet werden kann, wie für die Freigabe von Systemen für Arzneimittel,
Insektizide, Herbizide etc., als semipermeable Membranen bei der umgekehrten Osmose, als Körperimplantate und Verbandmaterial
oder als Material für Kontaktlinsen.
Hydrogele mit einer lederartigen Zähigkeit in trockenem Zustand und einer hohen Dehnung im nassen Zustand können in Abhängigkeit
von den relativen Anteilen der zuvor beschriebenen Komponenten mit einer Wasserabsorption, die von niedrig bis
hoch reicht, synthetisiert werden.
Hydrophile Membranen, die eine Naßfestigkeit bei relativ
hohem Gewicht mit Gleichgewichtswassergehalten von 5 bis 50 $
vereinen,sind besonders bei Anwendungen wie in Form von Körperimplantaten, Verbandmaterial und semipermeablen Membranen
verwendbar. Beispielsweise müssen subkutane und intramuskuläre Implantate für die kontrollierte Freigabe der
Medikation zu einer Wasserabsorption in einem mäßigen Bereich (15 bis 25 Gew.-^) befähigt sein, jedoch in trockenen Zustand
und gequollenem Zustand fest genug sein, um sowohl Einsatzais auch Extraktionsverfahren standzuhalten.
Zusätzlich zu ihrer Eignung als Arzneimittelträger sind die Hydrogele der vorliegenden Erfindung für die Verwendung als
Träger für Antiseptika, Geschmacksstoffe, färbende Kittel, Nahrungsmittel, Insektizide, Herbizide und dergleichen, geeignet.
Insbesondere können die erfindungsgemäßen Hydrogele in einem geeigneten Lösungsmittel, das das freizugebende Mittel

enthält, gequollen werden. Das Lösungsmittel wird verdampft,
wobei das Mittel innerhalb der Hydrogelteilchen zurückbleibt. Beim Kontakt mit einer wässrigen Umgebung wird das Mittel
in kontrollierter Weise freigegeben.
Aufgrund ihrer guten Gewebeverträglichkeit und Säuerstoffpermeabilität
zusätzlich zu einem weiten Bereich der Wasserabsorptionen und Festigkeit und Elastizität sind die erfindungsgemäßen
Hydrogele besonders für die Verwendung als intramuskuläre und subkutane Implantate bei warmblütigen
Lebewesen und als Kontaktlinsenmaterial verwendbar. Aus den gleichen Gründen können die erfindungsgeiräßen Hydrogelmaterialien
in Ersatzblutgefäße oder in Nebenschlüsse außerhalb des Körpers ohne die erforderliche Stützmatrix, die bei andersartigen
relativ schwachen Hydrogelmaterialien verwendet wird, übergeführt werden.
Die erfindungsgemäßen Hydrogelmaterialien sind auch für die
Verwendung als hydrophiler und nichtthrombogener Überzug geeignet, da sie stark an Glas, Metall und Kunststoff anhaften.
Aufgrund ihrer hohen Festigkeit und Elastizität ergeben sie starke Überzüge mit einer hohen Abriebsfestigkeit, die verwendbar
sind zum Überziehen von Bootsrümpfen, um einen Entenmus ehe Ib ewuchs abzuhalten, oder zum Überziehen von Linsen und
Glas, um ein Beschlagen in einer Umgebung mit hoher Feuchtigkeit zu verhindern. Aufgrund ihrer Hydrophilie in Verbindung
mit einer guten Sauerstoffpermeabilität sind sie auch als Überzüge für Samen geeignet.
Ein anderes einzigartiges Merkmal der erfindungsgemäßen Hydrogele ist ihre Flexibilität in trockenem Zustand, derart, daß
sie die bei der Anwendung gewünschte Form annehmen. Ein weiterer Vorteil der erfindungsgemäßen Hydrogele wird in gequollenem
Zustand in so weit verwirklicht, als sie nicht brüchig werden, sondern die Festigkeit und Elastizität, selbst
wenn sie sich in einem vollständigen Gleichgewicht in einer wässrigen Umgebung befinden, beibehalten. Diese Eigenschaften

sind besonders bei Membrananwendungen unter Druck wertvoll,
wie "bei einer Vorrichtung zur umgekehrten Osmose, für die die
erfindungsgemäßen Hydrogele geeignet sind. Zusätzlich zu
ihrer Zähigkeit und Flexibilität zeigen die erfindungsgemäßen
Polysiloxanhydrogele auch eine erhöhte Sauerstoffpermeabilität
die sie anderen in Frage kommenden Materialien für Kontaktlinsen überlegen machen.
Erfindungsgemäße Hydrogele, die Arzneimittel enthalten, sind
auch aufgrund ihrer flexiblen, sich der Form anpassenden Natur und ihrer gleichzeitigen Fähigkeit, Arzneimittel direkt
auf den in Mitleidenschaft gezogenen Bereich zu übertragen, besonders für die Verwendung bei der Behandlung von offenen
Wunden und Verbrennungen geeignet. Ein besonderer Vorteil der Hydrogele bei dieser Anwendung ist ihre Flexibilität in
trockenem Zustand. Es ist nicht erforderlich, das Hydrogel vor der Anwendung vorzuquellen, um es ausreichend weich und
geschmeidig zu machen für eine Umhüllung von großen, verletzten Flächen, und somit kann das das Arzneimittel enthaltende
Hydrogel eher in getrocknetem als in gequollenem Zustand gelagert werden, wodurch die Lagerfähigkeit des hierin
enthaltenen Arzneimittels erhöht wird.
Sämtliche Arten von Arzneimitteln, die für die Behandlung des Körpers verwendet werden, sowohl topische als auch systemische,
können als wirksamer Bestandteil in den Copolymerenträger der Erfindung eingebracht werden. Die Bezeichnung "Wirkstoff" wird
vorliegend in dem weitesten Sinn verwendet, derart, daß sie jegliche Zusammensetzung umfaßt, die eine pharmakologische
oder biologische Wirkung hervorruft.
Geeignete Arzneimittel für die Verwendung bei der Therapie
gemäß der Erfindung umfassen ohne Einschränkungen diejenigen,
die in der US-PS 3 732 865 (Spalten 10 und 11) angegeben sind.
Andere Arzneimittel mit der gleichen oder einer verschiedenen
physiologischen Aktivität wie die vorstehend zitierten, können

in Trägern innerhalb des Bereichs der Erfindung verwendet
werden. Geeignete Mischungen von Wirkstoffen können natürlich mit der gleichen Leichtigkeit wie bei aus einer einzigen Komponente
"bestehenden Systemen freigegeben werden.
Die Arzneimittel können in verschiedenartigen Formen, wie in Form von unbeladenen Molekülen, Komponenten von Molekülkomplexen
oder nicht irritierenden pharmakologisch annehmbaren
Salzen, wie das Hydrochlorid, Hydrobromid, Sulfat, Phosphat, Nitrat, Borat, Acetat, Maleat, Tartrat und Salicylat usw. vorliegen.
Für saure Arzneimittel können Salze von Metallen, Aminen oder organischen Kationen (z. B. quaternäres Ammonium) verwendet
werden. Weiterhin können einfache Derivate der Arzneimittel (wie Äther, Ester, Amide usw.), die die gewünschten Retentionsund
Freigabeeigenschaften besitzen, die jedoch durch den Körper-pH sowie Enzyme usw. leicht hydrolysiert werden, verwendet
werden.
Die Menge des in den Träger einzubringenden Arzneimittels variiert innerhalb eines weiten Bereichs in Abhängigkeit von
dem speziellen Arzneimittel, der gewünschten therapeutischen Wirkung und der Zeitspanne, während der das Arzneimittel
freigegeben werden soll. Da eine Tielzahl von Trägern mit einer Vielzahl von G-rößen und Gestalten vollständige
Dosierungssysteme für die Therapie von zahlreichen Krankheiten ergeben soll, gibt es keine kritische obere Grenze
hinsichtlich der Menge des in den Träger eingebrachten Arzneimittels. Die untere Grenze hängt auch von der Aktivität des
Arzneimittels und der Zeitspanne für seine Freigabe aus dem Träger ab. Somit ist es nicht zweckmäßig, einen Bereich für die
therapeutisch wirksame Menge des Arzneimittels, das durch den Träger freigegeben wird, zu definieren.
Zusätzlich zu den Arzneimitteln können in die erfindungsgemäßen
Copolymeren Duftstoffe oder Kahrungsmittelgeschmacksstoffe, wie Orangenöl, Citral, Kaffee, Tee, Citrusöl,
synthetischer Limonen-Citrusgeschmack, Erdbeergeschmack,

Vanille, Biacetyl, Anis, Veilchenduftstoff, Fichtenduftstoff,
Pfefferminzöl, Orchideenessenzöl, Anäthol, Äthylpropionat,
Äthylacetat, Acetaldehyd, Menthol und Krauseminze sowie Pestizide einschließlich von Bakteriziden, Fungiziden,
Insektiziden und Nematoziden eingebracht werden.
Weitere Beispiele für diese biologisch wirksamen Bestandteile sind in der ÜS-PS 3 660 563 (Spalten 3 bis 7) angegeben.
Die neuen Hydrogele stellen eine Mischung von drei Polymerentypen
dar, die die Wasserquellbarkeit der herkömmlichen HEMA-HydrogeIe, die Festigkeit der Polyurethane und die
biologische Inertheit der Polysiloxane in einem einzigen Polymeren vereinen. Somit sind sie besonders wertvoll als
biomedizinische Kunststoffe im Kontakt mit Blut und Gewebe und können als ausgezeichnete in Frage kommende Materialien
für synthetische Haut bei der Behandlung von Verbrennungen,als
künstliche Organe im Hinblick auf deren Konstruktion oder als Überzugsmaterial hierauf oder als Kahtmaterialien angesehen
werden.
Die Hydrogelzusammensetzungen können von 20 bis 90 $ Polymeres
(Ap) von dem Monomeren (A) und 10 bis 80 fo Siloxanmakromeren
(B), entsprechend einem typischen Grad der Quellung von ca. 10 bis 250 fo variieren. Die bevorzugten
Zusammensetzungen enthalten von ca. 15 bis 60 <?o Makromeres
(B) und ca. 40 bis 85 fo Polymeres (Ap) und besitzen einen
Bereich hinsichtlich ,ihres Quellungsgrades von 15 bis 120 $.
Die folgenden Beispiele erläutern die Erfindung. In diesen Beispielen wird der Quellungsgrad, DS, wie folgt definiert:

Die folgenden Beispiele erläutern die Erfindung. Beisp_iel_1_
Herstellung_des_Siloxanmakromeren
Man beschickte einen 1000 ml Dreihalskolben, der mit einem mechanischen Rührer, einem Thermometer, einem Kühler und
einem Stickstoffeinlaß versehen war, mit 480 g (0,2 Mol)
Polydimethylsiloxandiol, Q4-3667 der Dow Corning mit einem Molekulargewicht von 2400 und 88,8 g (0,4 Mol) Isophorondiisocyanat.
JTach der Zugabe von 0,2 g Triäthylaminkatalysator
wurde die Mischung unter Stickstoff "bei 60 C fünf Stunden gerührt, bis der Isocyanat (ΗΌΌ)-Gehalt 2,78 io
(durch Titration, 2,95 $ theoretisch) erreichte.
Man verdünnte 500 g dieses Polydimethylsiloxandiisocyanatprepolymeren
mit 125 g 2-Hydroxyäthylmethacrylat, um eine
87,3 /Sige lösung des Siloxanprepolymeren, das endständig mit
Methacrylatteilen (= Siloxanmakromeres) versehen war, in 2-Hydroxyäthylmethacrylat.
Diese Lösung wurde bei Raumtemperatur gerührt, bis die Isocyanat (ETCQ)-Bande aus den aus
der Lösung entnommenen Infrarotspektren verschwand.
Man fügte zu 100 g der in Beispiel 1 hergestellten Siloxanmakromerenlösung
0,1 g tert.-Butylperoctoat. Die Makromerenlösung in 2-Hydroxyäthylmethacrylat wurde 30 Min. entgast,
und dann zwischen "Teflon"-überzogene Aluminiumfolien, die
mit einem Siliconschnurabstandshalter mit einem Durchmesser von 1,588 mm (1/16 inch) versehen waren, gegossen. Die Foiienform
wurde bei 800C 3 Stunden mit einer einstündigen Nachhärtungsperiode
bei 1000C gehärtet. Fach dem Abkühlen erhielt man eine durchscheinende, sehr flexible Polymerenfolie mit
einer Dicke von 1,5 mm.

Unter Befolgung des Verfahrens von Beispiel 2 stellte man eine Folie her, indem man eine 43,7 $ige lösung des Siloxanmakromeren
in 2-Hydroxyäthylmethacrylat, hergestellt durch
Zugabe von 50 g der Makromerenlösung von Beispiel 1 zu 50 g 2-Hydroxyäthylmethacrylat, härtete.

Unter Befolgung des Verfahrens von Beispiel 2 stellte man eine Folie her, indem man eine 54,6 $ige Lösung des Siloxanmakromeren
in ^-Vinylpyrrolidon und 2-Hydroxyäthylmethacrylat,
hergestellt durch Mischen von 62,5 g der Makromerenlösung von Beispiel 1, 12,5 g 2-Hydroxyäthylmethacrylat und 25 g
N-Vinylpyrrolidon?härtete.

Unter Befolgung des Verfahrens von Beispiel 1 wurde ein
anderes Siloxanmakromeres hergestellt, indem man zuerst 160 g
(0,2 Mol) Polydimethylsiloxandiol, Q4-3557 der Dow Corning
mit einem Molekulargewicht von 800 und 88,8 g (0,4 Mol) Isophorondiisocyanat umsetzte. Die Mischung wurde 5 Stunden
bei 5O0C unter Stickstoff gerührt, bis der NCO-Gehalt 6,45 #

Unter Befolgung des Verfahrens von Beispiel 1 wurde ein weiteres Siloxanmakromeres hergestellt, indem man zuerst
180 g (0,03 Mol) Polydimethylsiloxantriol, 1248 der Dow Corning, mit einem Molekulargewicht von 6000 und 20 g
(0,09 Mol) Isophorondiisocyanat umsetzte. Die Mischung wurde 12 Stunden unter Stickstoff bei 800C gerührt, wonach 0,06 g
1,4-Diazabicyclo-[2,2,2 !-octan, "Dabco", als Katalysator
zugegeben wurden, bis der HOO-Gehalt 1,89 ^ (durch Titration,
1,89 $ theoretisch) erreichte.
Man verdünnte 170 g Siloxantriisocyanatprepolymeres mit 42,5 g 2-Hydroxyäthylmethacrylat und rührte bei Raumtemperatur
bis die NCO-Bande in dem Infrarotspektrum verschwand.

Unter Befolgung des allgemeinen Verfahrens von Beispiel 2
wurde eine Folie hergestellt, indem man eine Mischung von 45 g der Siloxanmakromerenlösung von Beispiel 8, 15g
N-Viny!pyrrolidon, 60 g Vinylacetat und 0,12 g Azobisisobutyronitrilkatalysator
härtete. Diese Mischung wurde 15 Min. entgast und dann zwischen "Teflon"-überzogene Aluminiumfolien
gegossen, die mit einem Siliconschnurabstandhalter mit einem Durchmesser von 1,5885 mm (1/16 inch) versehen war.
Die FoIienform wurde "bei 500C 16 Stunden mit einer einstündigen
iTachhärtungsperiode "bei 1000C gehärtet. Fach dem
Abkühlen wurde eine harte durchscheinende Folie erhalten.
Unter Befolgung des Verfahrens von Beispiel 1 wurde ein
weiteres Siloxanmakromeres hergestellt, indem man zuerst
150,3 g (0,05 Mol) Polydimethylsiloxandithiol, X2-8024 der Dow Corning, Molekulargewicht 3006, und 22,2 g (0,10 Mol)
Isophorondiisocyanat umsetzte. Fach der Zugabe von 0,06 g
Triäthylaminkatalysator wurde die Mischung 7 Stunden bei
500C unter Stickstoff gerührt, bis der NCO-Gehalt 2,72 $
(durch Titration, 2,43 $ theoretisch) erreichte.
Man verdünnte 100 g des Siloxandiisocyanatprepolymeren mit 25 g 2-Hydroxyäthylmethacrylat und rührte bei Raumtemperatur,
bis die HOO-Bande in dem Infrarotspektrum verschwand.

Unter Befolgung des Verfahrens von Beispiel 9 wurde eine Folie
hergestellt, indem man eine Mischung von 45 g der Siloxanmakromerenlösung von Beispiel 10, 15g ^-Vinylpyrrolidon,

60 g Vinylacetat und 0,12 g Bisisobutyronitrilkatalysator
härtete.
Die verschiedenen in den Beispielen 2 bis 4, 6, 7, 9 und 11
hergestellten Folien wurden hinsichtlich ihrer Zugfestigkeit und Dehnung sowohl in feuchtem als auch in naßem
(= gequollenem) Zustand im Gleichgewicht mit Wasser sowie auch hinsichtlich ihres Quellungsgrades sowohl in Wasser
als auch in Äthanol untersucht.
Die Ergebnisse dieser Messungen sind in Tabelle I angegeben. Der Quellungsgrad (DS) in Wasser oder Alkohol wird bestimmt,
indem man ein gegebenes Gewicht des Polymeren in Wasser oder Alkohol bis zur Einstellung des Gleichgewichts quillt, das
gequollene Polymere wiegt und die getrocknete Probe erneut wiegt. Der Quellungsgrad ist wie folgt definiert:

der Siloxanmakromerenlösung von Beispiel 5 und 12,5 g lT-Vinyl-2-pyrrolidon hergestellt.
Die verschiedenen in den Beispielen 12 bis 19 hergestellten
Folien wurden hinsichtlich ihrer Zugfestigkeit und Dehnung
sowohl in trockenem als auch naßem (= gequollenem) Zustand im Gleichgewicht mit Wasser sowie hinsichtlich des Quellungsgrades
sowohl in Wasser als auch in Äthanol untersucht. Die Ergebnisse dieser Messungen sind in Tabelle II angegeben.

Die folgenden Beispiele 20 bis 22 beschreiben die Synthese und die Bewertung von Filmen für Gasdiffusionsmessungen.
Beisp_iel_20
Man fügte zu 10 g der in Beispiel 4 hergestellten Siloxanmakromerenlösung
0,01 g tert.-Butylperoctoat. Die Makromerenlösung
wurde 30 Min. entgast und dann zwischen Mylar-ausgekleidete Glasplatten mit einem 0,127 mm (5 mil) dicken
Mylarabstandshalter gegossen. Die Folienform wurde 3 Stdn.
bei 8O0C mit einer einstündigen Fachhärtungsperiode bei
1000C gehärtet. Nach dem Abkühlen wurde eine klare flexible
Polymerenfolie mit einer Dicke von ca. 0,089 mm (3,5 mil) erhalten.
Unter Befolgung des Verfahrens von Beispiel 20 stellte man mit den Zusammensetzungen der Beispiele 12, 13, 16 und 18
Filme her. Man maß die Säuerstoffdiffusion und -permeabilität
in trockenem Zustand.

2. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das wasserlösliche Monomere ausgewählt ist unter Acrylsäure,

Imiden dieser Säuren, Salzen der monoolefinischen Sulfonsäuren und monoolefinischen monocyclischen azacyclischen Verbindungen.
3. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das wasserlösliche Monomere Acrylsäure, Methacrylsäure, ein
Hydroxyalkyl- oder Dialkylaminoalkylester dieser Säuren, worin Alkyl 2 bis 4 Kohlenstoffatome enthält, ist.
4. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das wasserlösliche Monomere ein Acrylsäure- oder Methacrylsäureester
eines Alkohols der Formel

5. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das wasserlösliche Monomere ein Amid oder Imid der Acryl- oder
Methacrylsäure ist, worin der N-Substituent Hydroxyalkyl, Oxaalkyl oder Dialkylaminoalkyl, worin Alkyl 2 bis 4 Kohlenstoff
atome aufweist, ist.
6. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das wasserlösliche Monomere 2-Hydroxyäthylacrylat, 2-Hydroxyäthylmethacrylat,
2-Hydroxypropyiacrylat, 2-Hydroxypropylmethacrylat,
3-Hydroxypropylacrylat, 3-Hydroxypropylmethacrylat, 2,3- Dihydroxypropylacrylat,
2,3-Dihydroxypropylmethacrylat oder N-Vinyl-2-pyrrolidon ist.

10. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das wasserlösliche Monomere ein HydroxyalkyIvinylather ist, worin
Alkyl 2 bis 4 Kohlenstoffatome aufweist.
11. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das wasserunlösliche Monomere ausgewählt ist unter den Alkylacrylaten
oder -methacrylaten, worin Alkyl 1 "bis 18 Kohlenstoff atome aufweist, Tinylestern der Alkansäuren mit 1 "bis
Kohlenstoffatomen, Vinylbenzoat, Acrylnitril, Styrol und Vinylalkyläthern, worin Alkyl 1 "bis 5 Kohlenstoff atome aufweist.
12. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das wasserunlösliche Monomere ein Alkylacrylat oder -methacrylat,
worin Alkyl 1 "bis 5 Kohlenstoff atome aufweist, Vinylacetat,
Styrol oder Acrylnitril ist.
13· Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß das
Siloxanmakromere ein Molekulargewicht von ca. 600 "bis ca.
8500 aufweist und ein Polysiloxanpolyol oder -dithiol mit einem Molekulargewicht von ca. 400 bis ca. 8000 umfaßt, das
zuerst mit Isophorondiisocyanat oder Tolylen-2,4-diisocyanat
endständig abgeschlossen und dann mit einer Hydroxyverbindung umgesetzt wurde, die ausgewählt ist unter den Hydroxyalky1-estern,
worin Alkyl 2 bis 4 Kohlenstoffatome aufweist, der

14· Gel gemäß Anspruch 13, dadurch gekennzeichnet, daß die
Hydroxyverbindung 2-Hydroxyäthylacrylat, 2-Hydroxyäthylmethacrylat,
2-Hydroxypropylacrylät, 2-Hydroxypropylmethacrylat,
3-Hydroxypropylacrylat, 3-Hydroxypropylmethacrylat, 2,3-Dihydroxypropylacrylat
oder 2,3-Dihydroxypropylmethacrylat ist.
15. Gel gemäß Anspruch 13, dadurch gekennzeichnet, daß die
Hydroxyverbindung 2-Hydroxyäthylacrylat oder 2-Hydroxyäthy1—
methacrylat ist.
16. Gel gemäß Anspruch 13, dadurch gekennzeichnet, daß das
Polysiloxanpolyol oder -dithiol endständig mit Isophorondiisocyanat
abgeschlossen ist und die Hydroxyverbindung 2-Hydroxyäthylmethacrylat ist.
17. Gel gemäß Anspruch 1, dadurch gekennzeichnet, daß ca. 15
bis ca. 60 Gew.-^ des Gels das Siloxanmakromere sind.
18. Verwendung des Gels gemäß Anspruch 1 als wasserunlöslicher
hydrophiler Bestandteil bei Formulierungen wie pharmazeutischen Zusammensetzungen, Duftzusammensetzungen, pesticiden
Zusammensetzungen, herbiciden Zusammensetzungen und in Kontaktlinsen.
