Abstract
Automatischer Analysator (50) zur Verwendung bei der Durchführung eines Nucleinsäure-basierten Assays, wobei der Analysator (50) in einem Gehäuse (60) umfasst:
eine Magnetbewegungsstruktur (810) zum Aussetzen einer Probe einem Magnetfeld, wobei die Magnetbewegungsstruktur (810) eine Vielzahl von Magneten (814) trägt;
ein Absaugrohr (860) zum Absaugen einer flüssigen Komponente der Probe, während die Probe dem Magnetfeld der Magnetbewegungsstruktur (810) ausgesetzt wird;
einen Amplifikationsinkubator (604), umfassend ein Gehäuse (610), das eine Aufnahmegefäßzugriffsöffnung (614) umfasst, die zum Bewegen eines Reaktionsaufnahmegefäßes (162) in den oder aus dem Amplifikationsinkubator (604) dimensioniert ist, wobei angenommen wird, dass das Reaktionsaufnahmegefäß (162) eine Target-Nucleinsäure abgetrennt von der flüssigen Komponente der Probe enthält, und
eine Detektionsvorrichtung (950) zum Detektieren eines Lichtsignals, das mit einem Amplifikationsprodukt, gebildet in dem Amplifikationsinkubator, assoziiert ist, wobei das Amplifikationsprodukt für das Vorliegen der Target-Nucleinsäure in der Probe indikativ ist.
eine Magnetbewegungsstruktur (810) zum Aussetzen einer Probe einem Magnetfeld, wobei die Magnetbewegungsstruktur (810) eine Vielzahl von Magneten (814) trägt;
ein Absaugrohr (860) zum Absaugen einer flüssigen Komponente der Probe, während die Probe dem Magnetfeld der Magnetbewegungsstruktur (810) ausgesetzt wird;
einen Amplifikationsinkubator (604), umfassend ein Gehäuse (610), das eine Aufnahmegefäßzugriffsöffnung (614) umfasst, die zum Bewegen eines Reaktionsaufnahmegefäßes (162) in den oder aus dem Amplifikationsinkubator (604) dimensioniert ist, wobei angenommen wird, dass das Reaktionsaufnahmegefäß (162) eine Target-Nucleinsäure abgetrennt von der flüssigen Komponente der Probe enthält, und
eine Detektionsvorrichtung (950) zum Detektieren eines Lichtsignals, das mit einem Amplifikationsprodukt, gebildet in dem Amplifikationsinkubator, assoziiert ist, wobei das Amplifikationsprodukt für das Vorliegen der Target-Nucleinsäure in der Probe indikativ ist.
Claims (14)
- Automatischer Analysator (
50 ) zur Verwendung bei der Durchführung eines Nucleinsäure-basierten Assays, wobei der Analysator (50 ) in einem Gehäuse (60 ) umfasst: eine Magnetbewegungsstruktur (810 ) zum Aussetzen einer Probe einem Magnetfeld, wobei die Magnetbewegungsstruktur (810 ) eine Vielzahl von Magneten (814 ) trägt; ein Absaugrohr (860 ) zum Absaugen einer flüssigen Komponente der Probe, während die Probe dem Magnetfeld der Magnetbewegungsstruktur (810 ) ausgesetzt wird; einen Amplifikationsinkubator (604 ), umfassend ein Gehäuse (610 ), das eine Aufnahmegefäßzugriffsöffnung (614 ) umfasst, die zum Bewegen eines Reaktionsaufnahmegefäßes (162 ) in den oder aus dem Amplifikationsinkubator (604 ) dimensioniert ist, wobei angenommen wird, dass das Reaktionsaufnahmegefäß (162 ) eine Target-Nucleinsäure abgetrennt von der flüssigen Komponente der Probe enthält, und eine Detektionsvorrichtung (950 ) zum Detektieren eines Lichtsignals, das mit einem Amplifikationsprodukt, gebildet in dem Amplifikationsinkubator, assoziiert ist, wobei das Amplifikationsprodukt für das Vorliegen der Target-Nucleinsäure in der Probe indikativ ist. - Automatischer Analysator (
50 ) nach Anspruch 1, wobei die Magnetbewegungsstruktur (810 ) zwischen einer ersten und einer zweiten Position bewegbar ist, wobei die Magnete (814 ) keine wesentliche Wirkung auf magnetisch ansprechende Partikel, die in der Probe vorliegen, haben, wenn die Magnetbewegungsstruktur (810 ) in der ersten Position ist, und wobei die Magnete (814 ) eine wesentliche Wirkung auf magnetische ansprechende Partikel, die in der Probe vorliegen, haben, wenn die Magnetbewegungsstruktur (810 ) in der zweiten Position ist. - Automatischer Analysator (
50 ) nach Anspruch 1 oder 2, wobei die Detektionsvorrichtung (950 ) ein Photomultiplier-Rohr (956 ) zum Detektieren des Lichtsignals umfasst. - Automatischer Analysator (
50 ) nach Anspruch 1 oder 2, wobei die Detektionsvorrichtung (950 ) einen optischen Sensor zum Detektieren des Lichtsignals umfasst. - Automatischer Analysator (
50 ) nach Anspruch 4, wobei der optische Sensor eine Diode ist. - Automatischer Analysator (
50 ) nach einem der Ansprüche 1 bis 5, außerdem umfassend eine Temperaturrampensteuerungsstation (700 ), die sich zwischen der Magnetbewegungsstruktur (810 ) und dem Amplifikationsinkubator (604 ) befindet, wobei die Temperaturrampensteuerungsstation (700 ) eine Heizvorrichtung oder einen Chiller zur Erhöhung oder Senkung der Temperatur einer Flüssigkeit, von der angenommen wird, dass sie die Target-Nucleinsäure enthält, umfasst. - Automatischer Analysator (
50 ) nach einem der Ansprüche 1 bis 6, außerdem umfassend einen Transportmechanismus (502 ) zum Bewegen des Reaktionsaufnahmegefäßes (160 ) in den und aus dem Amplifikationsinkubator (604 ). - Automatischer Analysator (
50 ) nach Anspruch 7, wobei der Transportmechanismus (502 ) einen ausziehbaren und einziehbaren Bedienungshaken (506 ) zum Eingreifen in das und zum Bewegen des Reaktionsaufnahmegefäßes (160 ) in den und aus dem Amplifikationsinkubator (604 ) hat. - Automatischer Analysator (
50 ) nach Anspruch 7 oder 8, wobei der Transportmechanismus (502 ) drehbar ist. - Automatischer Analysator (
50 ) nach einem der Ansprüche 1 bis 9, außerdem umfassend gelagerte Reagentien zur Durchführung einer Nucleinsäure-basierten Amplifikationsreaktion. - Verfahren zur Bestimmung des Vorliegens einer Target-Nucleinsäure in einer Probe, wobei das Verfahren Durchführen der automatisierten Schritte: (a) Kombinieren einer Probe mit einem festen Trägermaterial unter Bedingungen, die erlauben, dass die Target-Nucleinsäure an dem festen Trägermaterial immobilisiert wird; (b) Isolieren des festen Trägermaterials von nicht-immobilisierten Komponenten der Probe und Absaugen der nicht-immobilisierten Komponenten der Probe; (d) Waschen des festen Trägermaterials einmal oder mehrmals mit Waschpuffer; (e) Amplifizieren einer Targetsequenz, die in der Target-Nucleinsäure enthalten ist; und (f) Detektieren eines Amplifikationsprodukts, das in Schritt (e) gebildet wurde, als Hinweis für das Vorliegen der Target-Nucleinsäure in der Probe, in einem Gehäuse umfasst.
- Verfahren nach Anspruch 11, wobei das feste Trägermaterial ein magnetisch ansprechendes Partikel ist und wobei Schritt (b) Aussetzen des festen Trägermaterials einem Magnetfeld umfasst.
- Verfahren nach Anspruch 11 oder 12, wobei die Target-Nucleinsäure mit einer Fangsonde an dem festen Trägermaterial immobilisiert wird.
- Verfahren nach einem der Ansprüche 11 bis 13, wobei der Detektionsschritt ein Hybridisieren einer detektierbar markierten Sonde an das Amplifikationsprodukt umfasst.
