Abstract

连续波(CW)分光光度计已被广泛地用于在生物细胞组织中确定一个光吸收色料(例如血红素、氧化血红素)的自然条件下的集聚度。例如,此种分光光度计在脉冲血氧定量剂中将光线引入一个手指或耳垂,以测量光的衰减并随之根据比尔莱姆博特(Beer Lambert)公式或修正的比尔莱姆博特吸收公式来测定其集聚度。该比尔莱姆博特公式(1)描述了在吸收分量(C)、吸光系数(∈)、光入路长<L>和衰减光强度(I/Io)之间的关系加入公式(1)。
可是,直接使用比尔莱姆博特公式会引起一些问题。由于细胞组织的结构以及生理变化很大,所以入进光子的光路长也变化很大且不能由光源及检测器的几何位置简单而定。此外,光子入进路长的本身是吸收分量的相关集聚度的函数。举例来说,经过具有高的血液血红蛋白集聚度的器官的路长会不同于具有低的血液血红蛋白集聚度的同一器官的路长。而且,由于许多细胞组织构成的吸收系数依赖于波长,因而使路长时常依赖于光的波长。针对这一问题的一个结论是同时确定∈、C和<L>,但利用CW血氧定量计这一点是不可能的。
而且,对于小体积(例如手指)的细胞组织的定量测量来说,光子的逃逸会引入严重的误差,因为从该细胞组织逃逸掉的光子会被计为已经被吸收的光子。
存在有若干种原因需要使用体内细胞组织的血氧定量计。尽管可在自然条件下定量估计动脉氧饱和度,但不可能估计在当血液离开动脉并进入微血管基时在血红素氧的集聚度中的改变。而且,由于尚无可以用仪器所实现的技术用来从微血管基直接提取血样,所以也不可能确定从静脉回流在特定微血管基中的氧的饱合度。
在时间分解(TRS脉冲)和相位调制(PMS)分光光度计中,能够直接测量入进光子的平均路长,但只有当该光谱是以相当大的光源检测器分离度而被收集时才能执行时间分解或频率分解的频谱的正确的量化。对于象耳轮、手指或活体检查细胞组织这样的小体积的细胞组织来说,这种分离难以实现。
因此需要一种分光光度系统及其方法,用于定量检验相当小体积的生物细胞组织。
本发明实现一种分光光度系统,它旨在利用可见光或红外辐射来检验相当小体积的生物细胞组织。
根据本发明的一个方面,用于相当小的所感兴趣的被测物体(例如生物细胞组织、器官或处于固态、液态或气态的非器官物质)检测的分光光度系统利用可见或红外辐射使之进入贯穿经过该物体的路径。该系统包括一个分光光度计,具有一个用以把辐射引入到被测物体内的光输入端口和用以检测已入进物体之内经过一路径的辐射的一个光检测端口;环绕在该相当小所感兴趣被测物体放置的光子逃逸防止装置,用以禁止所引入的光子逃逸到被测物体的外部;以及处理装置,根据在引入的辐射和被检测的辐射之间的变化而用于确定被检测物体的光的特性。
依照本发明的另一方面,采用可见或红外辐射来检测所感兴趣的相当小的体积的生物细胞组织的一个系统包括一个分光光度计,在一光的输入端口处具有一个用来引入辐射的光源,一个采用来检测已入进经过从输入端口至一个光的检测端口的一个路径的辐射的检测器;以及一个采用来测定被引入和被检测的辐射之间的改变的一个处理器。该系统还包括一个相当大体积的光介质,形成一个防光子逃逸装置,具有可选的散射特性及吸收特性;采用来把所感光趣的生物细胞组织定位到入进路径中以产生一个细胞组织——介质光路的一个定位装置;该光介质实际上限制了光子从细胞组织——介质光路的逃逸;以及一个处理装置,采用来根据被检测的细胞组织——介质光路的光特性以及该光介质的散射或吸收特性而确定该细胞组织的生理特性。
本发明在这些方面的最佳实施例包括有下列特征之一或多个。

参考图1,用以检测相当小体积的生物细胞组织的系统10包括一个可选光学特性的光介质12、一个分光光度计18、滴定循环系统30和一个计算机控制部分35。所感兴趣的生物组织14被附在一个定位器15上,浸入于光介质12中。采用经光导20和22传导的可见或红外光,分光光度计18检测介质12的光特性。在最佳实施例中,光导20和22是由光纤构成,分别与光源21和光检测器23相连。在一光输入端口19引入的光子经一条散射及吸收路径入进介质12,并在检测端口被检测。输入端口19和检测端口21的可选的固定几何构形控制了入进路径,即光场25。
系统30被用来精确地改变介质12的散射和吸收特性、介质12包括根据其集聚度而展示为散射特性的脂内类溶液(由地处clapton,北卡罗莱纳州的Kabi Vitrum公司制造)以及展示为吸收特性的炭黑墨汁。介质12的散射及吸收特性可以通过适当溶液的混合而被保持恒定及均匀或通过在滴定系统30中构成成分的集聚度的改变而被几乎连续地改变。管道32和34用于这种溶液的连续循环。
在系统的操作中,细胞组织14一开始被置于远离光场25。分光光度计18检测在场区25中的介质12,而控制部分35将所检测的数据与吸收系数(μa)和散射系数(μs)的预选值相比较。随后,定位器15将细胞组织14定位于场25中,而分光光度计18测量组织14和介质12的光特性。根据所采集的具有及不具有细胞组织14的光谱数据,计算机控制部分35确定细胞组织14的光特性。
在另一个最佳操作方法中,在测量介质12的光特性之后,通过滴定而使介质12的散射和吸收特性匹配至组织14的特性,从而使得当该组织被插入到场25中时,其细胞组织14不引起场25的波动。在把介质12的散射和吸收系数匹配至细胞组织14的系数之后,分光光度计18检测具有和不具有细胞组织14的同一数据。已知的μa *和Us *的滴定值等于细胞组织μa和Us值。该匹配过程首先是匹配μa而后是μs,反之亦然。
所描述的方法既可应用于体内的组织检验也可应用于试管内的细胞组织的检验。细胞组织14可以是由透光材料所封闭的活体检验的样本,或者是插入介质12的人手指的一部分。由分光光度计18所使用的光的波长依照所感兴趣的细胞组织成分而选定(例如血红蛋白、氧化血红蛋白、葡萄糖、酶);采用多种波长也是在本发明的范围内。
本发明也包括光介质12的不同最佳实施例的采用。参考图2,填充有介质12的中空筒42环绕着手指40并防止引入光子的逃逸。介质12的光的特性、压力受到经管道32和34连接到圆筒42的系统30的控制。圆筒42的内壁是由柔韧、阻止光透材料44所构成。在手指插入该圆筒42内之后,该光阻物44充满地挤压在手指周围。该内部光阻物44的尺寸要使得在手指40抽出之后其介质12完全填充圆筒42的体积。这使得介质12的背景测量以及在介质12中的手指40的测量都以与图1相关描述的同一方式执行。受输入端口19和检测端口21的位置所控制的光场25或是几何透射型或是几何反射型。
参考图2B,在另一个实施例中,圆筒42由一套圆筒42A、42B、42C……所取代,每一个都包括具有恒定预选吸收和散射系数的流体或固体的介质12。固体光介质是钛或其它散射材料,嵌入在吸收性柔韧介质中,例如镉。
人的手指被插入每一个圆筒中,并由分光光度计18对插入的手指的光特性进行测量。利用圆筒和输入端口——检测端口的几何状的已知的光特性,其手指的光特性(即μa和μs)可与这些圆筒之一的光特性相匹配。
分光光度计18的最佳实施例是一个连续波分光计、一个相位调制分光计和一个时间分解分光计,它们在上述所引证的文献中均有讨论。
以双重波长的连续波分光计工作的系统10被用作一个手指血氧定量计。如图2A所示,由环绕的介质12防止了引入手指40内的光子的大部分的逃逸。因此,引入手指的光子或者是被吸收,或者是达到检测端口21并由检测器所接收。由于被吸收光子的出现,没有计数逃逸的光子的误差。对应于圆筒42的μa *和μs *的每一个所选值的背景光谱数据被存储在系统中,以便能够针对每一个波长与手指及圆筒的μa和μs的值相匹配。对于工作在敏感于血红素(Hb)和氧化血红素(HbO)的两个波长(例如754nm和816nm)的分光计来说,该血红素饱合度(Y)是以吸收系数的比率并按照以下用于氧的饱合度的等式来计算的:
其中该系数是从血红素在754nm和814nm的衰减值而确定的,该衰减值分别是εHb=0.38cm-1mM-1,εHb=0.18cm-1mM-1;而且在氧化血红素和血红素之间的差异衰减系数分别是ΔεHbO-Hb=0.025cm-1mM-1和ΔεHbO-Hb=0.03cm-1mM-1。
如本专业技术人员所公知,在血红素饱合度的测量中,血氧定量计规化了被检测到的数据以消除由于血液体积的改变而引起的偏差。但是,该体积的改变可被用于检测脉冲速率。
此外,通过在距检测器若干厘米处测量所引入的正弦调制光的强度和相位移θ,一个相位调制分光计被用于测量光子的入进。对于小体积的细胞组织,利用光介质12来实现在输入端口和辐射端口之间的最优距离。而且,介质12实际上消除了光子的逃逸。
被检测的相移直接与图2A所示的光子路径长度的分布的均值相关。光子入进的理论预言,被检测的光子在反射几何状方面将呈三维的“香焦型状”的分布图案,或在透射几何状方面为“雪茄型状”的分布图案。如果细胞组织的吸收特性不同于介质12的特性,将细胞组织14插入场25的中心就会引起路径长度分布的不均匀,即香焦型状的光场25是不均匀的。如果细胞组织的μa小于所环绕介质的μa则由于具有更长路径长度的光子将被更多地被吸收而使得平均路径长度<L>降低,反之亦然。因此,细胞组织14将引起在路径长度以及相位移θ方面的改变。
而且,被检测到的强度提供了一个调制指数(M),这是一个强散射介质的吸收和散射特性的一个重要的度量。该调制指数被确定为AC幅度(Aλ)对AC和DC的取和(DCλ)幅度的比率。
如Sevick等人在《分析化学》(卷195第330至351页,1991年。此文在此引作参考)中所描述,对于低的调制频率(即,2πf<<μaC),相移是渡越平均时间<t>的一个直接度量,即θ→2πf<t>。如果在一个介质中的全部光子以一恒定的速度C运行,则表示这种效果的相移,即平均路径长度θ→2πf<L>/C。在此,全部路径长度被加权相等。所确定的路径长度被用于比尔—莱姆勃特公式中以进行吸收特性的确定。
随着调制频率的增加,越短的路径长度将被更重地加权。在这些频率之处(即2πf>>μaC),相移后再是路径长度的分布的好的度量,而是正比于吸收系数μa和有效散射系数(1—g)·μs
由于有效散射系数于波长无关,在两个波长上所测的相移比率可被表示成
其中,θ0 λ是由散射及背景吸收所引发的在被测波长处的相移。该吸收系数的比率被用于细胞组织饱合度Y的确定。通过消除θ0,双频率、双波长相位调制分光计可被用于确定饱合度。吸收系数的比率被表示为在不同频率和波长处的相移的一个函数 在另一个最佳实施例中,一个时间——分解分光计(TRS—脉冲)在输入端口19处以小于微微秒的数量级输入光脉冲。穿行通过入进路径一个分布25的光子在检测端口21被收集。通过以接近无限边界条件、在一无限介质中求解作为格林(Green)函数的扩散方程式,从而确定在反射几何形R(ρt)(或透射几何形T(ρt))的被测光的强度。
由于在反射几何形中的半无限介质条件,该输入和输出端口的分离必须是在几个厘米的数量级以使用下面公式。 对于t→∞,吸收系数μa被确定为
其中ρ是输入和检测端口间的分离,C是在介质中的光速。有效的散射系数(1—g)μs被确定为
其中tmax是延时时间,在该延时处,被检测的反射时间的分布图(R(ρ1t)≡I(t))达到其最大值。公式7的左侧是修正过的脉冲到达时间的衰减陡度。由公式7所表明,通过测定所检测脉中的衰减陡度而对吸收系数量化。有效的散射系数(1—g)·μs由公式8所确定。对于已知的μa和μs以及输入端口、输出端口的几何性质,系统具有唯一的时间分布图I(t)。存储的分布图与被检测到的用于引入细胞组织的时间分布图相比较,以获得处理细胞组织14的散射和吸收系数的一个差异分布图。此外,通过改变介质12的散射和吸收特性而使介质12与细胞组织14的μa和μs相匹配,以使检测到的时间分布图不因引入细胞组织14而被改变。
TRS系统可被用于校准一个CW血氧定量计一定量化被测数据。为计数输入端口与检测端间的几何距离(ρ)和路径长度(<L>)之间的差异,某些血氧定量计采用如下所示的具有一差分路径长度因数(DPF)的经修正的比尔莱姆博特公式:

然而,CW血氧定量计不能精确地确定差分路径长度因数,因为该因数取决于路径长度。TRS按照下式利用吸收系数μa和散射系数μs来确定DPF:
用于新生儿头部(46)检测的防止逃逸光介质的一个可用实施例是一个光导发光极(optrode)支座45,如图所示。光纤20和22插入固体散射材料(例如泡沫聚苯乙烯)47中,它为逃逸光子48提供了回转路由。如图中的弯曲线箭头48所示,由于光子是经由散射材料返回细胞组织,则入进光子在细胞组织中的路径长度要长得多。因此,香焦形状的图案则将穿透得更深,并可用较小的输入——输出光纤的分离获得有意义的分光计数据而不会有由于实际上为直接路由所引起的光子逸漏或“短路”的危险。
系统10的不同的实施例可被用于执行所选生物特性的单一测量或连续、时间相关的监测。可以附加用于被测值连续监测的可视显示装置。而且,当被测值等于一预选值时可发出报警信号。
参考图4,在一项测试研究中,TRS—脉冲分光光度计被用于量化测定人的手指的散射和吸收特性。为创造半无限边界条件,被检测参量手指40被浸没入相当大体积的、具有含墨汁的炭的脂内类溶液52中。将购得的大约为20%集聚度的脂内类浮液稀释成大约0.5%—2.5%的集聚度,以产生环绕介质52。这种脂内类溶液的集聚度确定了该溶液的散射特性,而墨汁量控制着吸收特性。稀释炭黑墨汁的所选量按照需要加入到匹配介质中。在测试中,直径约15cm、高约8cm而体积约为9.4升的圆筒溶器51被用来容纳匹配介质52。几乎全部测试是针对25个健康志愿者(包括男性和女性)的参数手指而执行的,这些人中有白种人、亚洲人、非洲人。光纤56和60的光纤末端57和59被插入到主介质的溶液表面几个毫米之下,并被保持在被检测手指40两侧有3cm的间隔。手指40的插入方式是使其在环绕介质52表面的5—6cm之下而处在由浸入末端57和53所定义的光场54之内。这就避免了多数光子被透射到表面。
具有5MHz重复速率的双重波长TRS系统将在脉冲源62中所产生的红(670nm)或近红外(750和830nm)的100ps脉冲注入到介质52中。所使用的1μm直径的输入光纤56和2mm直径的输出光纤。检测器包括有连接到一个恒定分量识别器(CFD)66的具有150ps的时间分解度的一个微通道极板光倍增管64(MCP—PMT)。单一光子计数系统包括有一个时间——幅度转换器(TAC)68和用于寄存数字化数据的计算机70。在存在及不存在手指40时,都进行TRS的测量。
通过添加稀释的黑墨汁而首先提高环绕介质52的吸收系数μa(h)来采用上述的匹配方法。一旦确定了合适的吸收器的集聚度,就通过提高脂内类浮液的集聚度来采用第二滴定过程以确定μ′s(h)0。
利用补偿仪器响应的仪器函数对TRS数据进行反卷积。μa、μ′s及T0(即激光脉冲注入时间)被最小平方设置。吸收系数μa和散射系数μ′s是以log10为基底表示的,可以通过乘以2.303而转成以loge为基底表示。(注意:对于以方程式9计算的μs,这种转换不能采用。)
图4A示出了采用匹配方法和直接测量分别以670nm波长和2.5cm光纤间距对14人(4个白种人、5个亚洲人和5个非洲人)进行测试而获的吸收系数。在匹配测量中所获μa的相关值的变化范围是从0.05cm-1到0.08cm-1,很显然在三种人中的随机性;然而,在直接测量方面变化更大。与匹配方法相比,直接测量会获得高得多的μa值,这可能是由于当光纤被直接附在被测手指的表面时,其光子从手指的表面逃逸所引起的。图4B示出了对不同组志愿者所测的吸收值。图4C示出了作为观察数目的函数μa的值。在本项研究中,没有发现手指直径和吸收系数μa之间的关系,表明手指的大小对μa无影响。
图4D示出针对图4A的14个人以匹配方法在670nm测得的μs。散射数据在图4E中被总结为关于μs出现的函数。均值是6.62cm-1而误差是0.64cm-1,具有近似的高斯分布。
量化的手指血红素饱合度是以670nm和750nm测量的。由于在750nm处的水的吸收性起的作用相对地高,因而有必要从所计算的μa值中减去水的吸收背景。为此目的,我们假设水在750nm和670nm的吸收系数分别等于0.0041/cm和0.0261/cm。μa的背景校正值和相应的血红素饱合度值在下表中给出并在图4F和4G中给出。

Claims (30)
1、利用引入到通过所感兴趣的被测物体的光路中的可见或红外辐射检测该被测物的一种分光光度方法,其特征在于该方法包括以下步骤:
(a)环绕所说的被测物体提供一个光学装置,用于限制光子逃逸到所说被测物体的外部,
(b)在一光输入端口处将可见或红外范围内的所选波长的电磁辐射输入进该被测物体,
(c)检测已经入进所说被测物体从所说输入端口至一个光检测端口的辐射,
(d)根据引入辐射和被检测辐射之间的变化来确定所说被测物的光学性质。
2、根据权利要求1的分光光度方法,其特征在于,其中所说的用于限制光子逃逸的光学装置包括环绕所说被测物体、具有可选光特性的光介质。
3、根据权利要求2的分光光度方法,其特征在于,其中所说的可选光特性是一个吸收系数或一个散射系数。
4、根据权利要求1的分光光度方法,其特征在于,其中所说的用于限制光子逃逸的光学装置包括环绕所说被测物体的一个光介质,所说的光介质至少具有实际上与所说被测物的光学特性相匹配的一个光学特性。
5、根据权利要求4的分光光度方法,其特征在于,其中所说的光特性是一个吸收系数或一个散射系数。
6、根据权利要求1的分光光度方法,其特征在于,其中所说的确定步骤(d)包括:(e)选择包括一光介质的所说光学装置,具有与所说被测物体光学特性相近似的一个光学特性,
(f)执行所说的步骤(b)和(c)以测量所说被测物体的光特性,
(g)选择另一个光学装置,包括一个所说环绕介质其光学特性更接近于匹配所说被测物体的光学特性。
(h)交替地重复所说的步骤(f)和(g),直到所说环绕介质的光特性实质上与所说被测物体的光特性相匹配,
7、利用可见或红外辐射检测所感兴趣的相当小的生物细胞组织的一种分光光度方法,其特征在于,所说的方法包括以下步骤:
(a)提供具有可选散射和吸收特性的相当大体积的光介质,
(b)将所感兴趣的生物细胞组织引入到该介质内,使占据在一光输入端口和一光检测端口之间的一条光路的一部分并产生一个细胞组织——介质光路,
(c)在处于所说介质内的所说输入端口处引入在可见或红外辐射范围内的所选波长电磁辐射,
(d)在处于所说介质内的所说检测端口处检测已经入进通过所说细胞组织——介质光路的辐射,
(e)通过将被检测辐射的值与所说选定散射和吸收特性的所说介质对应的辐射值相比较来确定所说生物组织的吸收或散射特性,以及
(f)根据所说细胞组织的所说的确定的散射和吸收特性来检测所说细胞组织的生理特性。
8、利用可见或红外辐射检测所感兴趣的相当小的生物细胞组织的一种分光光度方法,其特征在于,所说的方法包括以下步骤:
(a)提供具有可选散射和吸收特性的相当大体积的光介质,
(b)在处于所说介质内的所说输入端口处引入在可见或红外辐射范围内的所选波长辐射,
(c)在处于所说介质内的所说检测端口处检测已经入进通过在所说介质内从输入端口开始的一条路径,
(d)从所说被测的辐射确定所说路径的散射或吸收特性,
(e)将所感兴趣的生物细胞组织引入到该介质内,使占据一条光路的一部分并产生一个细胞组织——介质光路,
(f)重复步骤(b)、(c)、(d)确定所说细胞组织——介质路径的散射或吸收特性,以及
(g)根据所说光路径和所说细胞组织——光介质路径之间的散射和吸收特性差异来检验所说细胞组织的生理特性。
9、利用可见或红外辐射检测所感兴趣的相当小的生物细胞组织的一种分光光度方法,其特征在于,所说的方法包括以下步骤:
(a)提供具有可选散射和吸收特性的相当大体积的光介质,
(b)将所感兴趣的生物细胞组织引入到该介质内,使占据在一光输入端口和一光检测端口之间的一条光路的一部分并产生一个细胞组织——介质光路,
(c)在所说介质中的所说输入端口处引入可见或红外辐射所选波长的、具有小于ns(10-9秒)数量级的持续期的脉冲,
(d)在所说介质中的所说检测端口处检测已经入进通过所说细胞组织——介质路径的辐射的渡过时间,
(e)相对于输入脉冲波形,以所说波长确定被检测脉冲波形的形状的改变,
(f)通过比较变化的波形和所选散射和吸收特性的所说介质的对应波形确定所说细胞组织的散射和吸收特性,和
(g)根据所说被确定的散射和吸收特性来检验所说细胞组织的生理特性。
10、利用可见或红外辐射检测所感兴趣的相当小的生物细胞组织的一种分光光度方法,其特征在于,所说的方法包括以下步骤:
(a)提供具有可选散射和吸收特性的相当大体积的光介质,
(b)将所感兴趣的生物细胞组织引入到该介质内,使占据在一光输入端口和一光检测端口之间的一条光路的一部分并产生一个细胞组织——介质光路,
(c)在处于所说介质内的所说输入端口处引入在可见或红外辐射范围内的所选波长电磁信号,所说信号已由能够确定所说光路径的一个平均长度的载波波形所调制,
(d)在处于所说介质内的所说检测端口处检测已经入进通过所说细胞组织——介质光路的辐射,
(e)将引入信号和被检测信号作比较,并从而确定该被检测信号对于该引入信号的相位移,所说的相位移是该细胞组织——介质路径的散射和吸收特性的指示,
(f)根据被检测的相位移和对应于选定散射和吸收特性的所说介质的相位移来确定所说细胞组织的散射和吸收特性,和
(g)根据所说细胞组织的所说的确定的散射和吸收特性来检测所说细胞组织的生理特性。
11、根据权利要求7、8、9、10的方法,其特征在于,进一步包括将已知改变引入所说介质的散射或吸收特性的步骤,直到所说细胞组织——介质路径的被测散射或吸收特性与所说介质的相应在特性匹配为止。
12、根据权利要求11的方法,其特征在于,进一步包括在确定所说生理特性过程中采用所说改变的已知值的步骤。
13、根据权利要求7、8、9或10的方法,其特征在于,其中所说的生理特性是血红素饱和度。

一个分光光度计单元,包括用以将辐射引入被测物体的一个光输入端口和用以测量已经入进该被测物体内的光路的辐射的一个光检测端口,
光子逃逸防止装置,环绕在所感兴趣的被测物,用以限制所引入的光子的逃逸到被物体外部,
处理装置,用以根据所引入的辐射和被检测的辐射之间的变化来确定被测物体的光学性质。
