Description

一种尼古丁吸入器、用于尼古丁 吸入器的芯管及其制备方法
本发明涉及一种尼古丁不能透过的容器及制造这种容器的方法。本发明的一个优选实施例是一种尼古丁吸入装置,这种装置容许使用者用口摄取尼古丁气化物并主要被用作戒烟辅助用具。
近年来越来越多的论据证明吸卷烟与诸如高血压和肺癌等疾病有关。美国1988年的卫生局医务主任报告(U.S.Surgeon General′sreport)中关于吸烟对健康的后果估计仅在美国每年就有300000人死于与吸烟有关的疾病。确实,过量的吸烟目前已在全世界范围内被认为是主要的健康疑难问题之一。
由于尼古丁的能成瘾的特性,瘾大的吸烟者要完全戒绝吸烟是一桩极端困难的事。虽然尼古丁是烟草烟雾中的危险因素之一,但在烟草燃烧中产生的其他物质,例如一氧化碳,焦油产物,醛类和氢氰酸被认为是比尼古丁更为严重的危险因素。
由于尼古丁的能成瘾的特性,一种可以被接受的吸烟替换方案是以吸烟以外的形式或方式来提供尼古丁。已经研制出有好几种能够完成此目的的产品。用作吸烟替代物和/或戒烟辅助用具的最成功的产品是称为Nicorette的一种橡皮口香糖,糖内含有尼古丁作为活性组分之一。这一产品是到目前为止为食物和药品管理局所认可的唯一形式的尼古丁替换品。
在这种橡皮口香糖内,尼古丁是以一种带有一种分布在橡胶基底内的不溶性阴离子交换剂(polacrilex)的络合物形式存在的。在此混合物中还包括一种缓冲剂。关于这一产品的有美国专利3877486;3901248;和3845217号。
在此领域研制出的另外一种产品是一种以“Favor”作为商标在美国市场上出现18个月的无烟香烟。此产品随后被取缔,国为它不符合食物和药品管理局的要求。这种产品的各种实施例在美国专利第4284089;4800903;和4813437号中有描述。
这种产品概括地说是使尼古丁能通过一根细长的管子来吸取,管子内装有一个含有尼古丁游离碱的海绵状聚合物存贮体。由吸用者吸气造成的一股气流将尼古丁气化物送入吸用者的肺中以满足其对尼古丁的需要。
在这种产品的具体商业实施例中,管子用聚对苯二甲酸丁二酯(PBTP)和聚乙烯(PE)聚合物制成。此管子用聚乙烯封套裹住以使尼古丁与外界隔离。但是出乎意外地发现尼古丁游离碱仍通过包装材料徙移出来并很快地从装置中逸走,这是因为尼古丁具有比预计更大的挥发性。据估计,未经冷藏的气化吸入物的贮存寿命大约为一个月。
本发明涉及的是对这种装置尼古丁游离碱的容器的改进,从而改进被贮藏的尼古丁的贮存期限及其纯度。
为解决上述的问题,在本发明的一个优选的实施例中,以芯管形式作为尼古丁吸入器的容器包括一个芯管外壳和一个在外壳内的通道,通道内安置有一个尼古丁存贮体。存贮体设计成能盛装一定量的尼古丁,其形式能容许尼古丁气化物释放在绕径或通过存贮体的流体流中。通道至少有两个与外壳的外面相通的开口以容许流体流穿过通道。存贮体由一个尼古丁不能透过的阻挡体将其与外界隔绝,阻挡体包括多个通道阻挡体部分,用于将通道在存贮体的两侧封住,而至少这些阻挡体部分是可以穿透的以使通道与外界相通。
在本发明的实施例中,芯管是一个圆筒,通道由圆筒的内壁所限定而在两端开口。尼古丁存贮体可以是一种聚合物塞头的形式,其内加入有尼古丁游离碱。为了使存贮体与外界隔绝,管子或圆筒可以用尼古丁不能透过的材料,例如一种丙烯腈和丙烯酸甲酯的共聚物制成。这种材料的一个实例是由B.P.-Sohio制造的商业名称为Barex的产品。
圆筒上的开口由一层薄的铝箔或其它型式的柔韧的、可穿透的、尼古丁不能透过的材料来密封。一种将铝箔密封于圆筒两端的简易方法是铝箔的内表面上可以涂一层薄的Barex涂层或薄膜,此合成物热封于圆筒的两端以形成通道阻挡体部分。
为了保护存贮体内的尼古丁在氧气存在的情况下不致变质,存贮体可以在没有氧气的环境中插进管内,并充入一种如氮那样的惰性气体。一种达到这一效果的方法是在氮的环境中向管内的尼古丁存贮体装料,然后将涂有Barex的沿箔密封于管子的两端。之所以选择Barex和铝作为材料是因为它们在贮存期限内出现的尼古丁渗透是微不足道的,而Barex又是一种良好的热封材料。
当吸入器准备应用时,可以把它放入一个专门设计的烟嘴内,烟嘴具有一个包围着通道的接受端,还设置有一个靠近烟嘴内通道的锋利尖头,用于当圆筒插入到接受端时刺穿圆筒的一端。圆筒的另一端可以用任何合适的用具,例如一种专门设计套装在管的另一端,还带有一个在通向外界的开口周围的锐利尖头的刀或柄状物的尖锐器。当芯管插进烟嘴内,其两端都被刺穿后,吸用者就可以用烟嘴吮吸,获得足够剂量的尼古丁气化物以满足他或她的烟瘾。
芯管可以装在分配箱中出售,箱内装有多支芯管连同一单个烟嘴。在芯管的外端需要用吸入器的部件以外的东西刺穿的实例中,该分配容器上还需包括一个尖刃以利于使用。
本发明也可以用于其它具体装置中,其中将尼古丁在使用前长期保存在一种容器中又能被使用者很容易取到。

本发明的实施例将参照附图予以详细说明。图1示出本发明的芯管10,它由一个圆筒体1 2形成,圆筒体形成一通道14,一股流体,例如空气,能够流过该通道。一个用于容纳尼游离碱的存贮体16安置在通道14中,其理由将在下面予以讨论。存贮体16可以由一个海棉体聚合物塞头形成,或者用其它的合适材料,如美国专利第4284089;4800903和4813437号中所描述的那些材料形成,这些专利的内容通过引用犹如在此全文公开式地结合在本说明书中。这三件专利目前与本申请中描述的发明为同一个单位所拥有。
为了所描述的本发明的目的,存贮体由海棉状聚乙烯形成,其中分布有一薄层液体尼古丁。海绵状塞头及其操作的细节以及尼古丁的成份在美国专利第4800903号中有详细叙述。
为本发明的目的,聚乙烯塞头内可以加入一种尼古丁,薄膜醇和乙醇的混合物。尼古丁与薄荷醇及乙醇三者的重量比最好为大约10∶1∶120。10∶1∶160的重量比经另外测试也证明有良好的功能。作为一个实例,用于大约150000个聚乙烯塞头的加进去溶液的混合物可以包括18000克乙醇,1500克尼古丁和150克薄荷醇。将给定数量的乙醇放在一个混合用容器(未显示)内,然后加入薄荷醇,不断地搅拌直等全部溶解为止。
然后将尼古丁加入溶液中,用手搅动大约三分钟。接着将一个密实的接合盖放在混合用容器上。冷凝器(未显示)中冷却水的温度调整至14℃并以10升/分钟的容量循环。一个内容量为260升的套层真空干燥器(未显示)的套层中的水以20±1℃的温度,5升/分钟的容量循环。将塞头放入真空干燥器中,并将容器抽空至小于27英寸水银柱。
尼古丁/乙醇溶液借助于负压吸入真空干燥器中,然后将真空阀关闭。真空度应当小于20英寸水银柱。真空干燥器于是以每分钟4转的速度旋转10分钟。然后将真空泵起动并打开真空阀,进入真空干燥器的水温提高到40±1℃。真空干燥器和泵应当一直开动到真空干燥器内部温度与进入该干燥器的水温之间的温差达到5-6°时为止。一种Kinney高真空泵型号KC-8曾被用于上述的操作过程中。
当达到上述的温差后,真空干燥器和泵就都停止运行。然后使真空干燥器中充满氮,并将聚乙烯塞头卸入一个专门设计的容器内,容器被抽空至负28英寸水银柱的压力,然后重新充以氮。这一程序连续重复进行直至所有的氧确已从装置中排出为止。接着将充氮的聚乙烯塞头装在一个充满氮的大容积容器内以保护尼古丁不受氧的侵袭。塞头然后在氮的环境中插进合适的管中,并以下述的方法予以密封。
在芯管10制成后为了防止氧渗入芯管10,及防止尼古丁逸出芯管10,圆筒体12是由尼古丁不能透过的材料制成的。用于此目的的一种合适材料是丙烯腈和甲基丙烯酸酯的共聚物,这种材料由B.P.-Sohio制造,商品名称为Barex。
已测试过许多种不同的化合物作为尼古丁不能透过的材料之用。开始时认为结晶聚合物由于其间隙空间小的特性可以作为一种好的选择物。但是发现这些化合物在阻止尼古丁徙移方面效力很小。意想不到的是Barex虽然是一种无定形的聚合物,却是一种有效的材料。
Barex特别适用于所述的这一用途,因为它是可热封的,可以在封  口处形成一个尼古丁不能透过的阻挡体,而且它的组成成分根据F.D.A.Regulation 21 CFR 175.105的规定可以允许用作粘接剂。Barex也可以粘附在铝或其它金属箔上,因而一种合适的、尼古丁不能透过的密封装置就能很容易地以铝箔为基层用热封法贴上一层或多层毗连的Barex膜而形成。
对于图1中的实施例,为了在存贮体16被插入后保存管内的惰性气体,管的两端用一层象铝箔18那样的尼古丁不能透过的阻挡体予以封住。此箔层通过一层粘在箔18上的Barex 20密封在Barex管12上,因而箔层18可以很容易地通过加热密封在Barex管12的两端。虽然Barex是用合适的粘接剂粘在铝箔上的,但是这种粘接剂不能用来将Barex层封接在一起或者将铝膜封接在Barex上,因为这种粘接剂本身并非是尼古丁不能透过的,而尼古丁就能通过封接件本身徙移出来。
如上面所述类型的芯管10可以与图2中所示的烟嘴2 2结合使用。通过形成用Barex制的圆筒体12和用涂有Barex的铝箔片料形成通道阻挡件部分,装进存贮体16内的尼古丁游离碱在含有尼古丁的存贮体16被插入并维持在一个无氧的环境中时就能防止其徙移出芯管10之外。例如,在芯管10中充以一种象氮那样的惰性气体,就能防止尼古丁游离碱由于与氧的相互作用而变质。以这种方式,吸用者在如下所述那样刺穿箔件18后就能吸取完全有效剂量的尼古丁。
上面所述结构的另一种形式是尼古丁不能透过的阻挡体可以用其它的方式形成。例如,管可以用PE或者其它类型的坚硬材料形成而在管的内表面粘上一层Barex。如果不用管,存贮体可以在任一个端上开有小孔,而整个存贮体全部涂上一层Barex,其两端都能如所讨论那样被刺穿。其它合适的芯管也能根据本发明形成,只要用尼古丁不能透过的阻挡体将尼古丁与外界隔绝,并且该阻挡体又能在需用时被刺穿以释放出尼古丁。
如图2所示,所用的烟嘴22包括一个口衔部分24和一个芯管夹持器26。一条通道28从口衔部分24延伸贯穿芯管夹持器2 6。
如图3A和3B所示,为了将芯管10装在烟嘴22内,将芯管10放在芯管夹持器26的外端处,靠近绕在通向芯管夹持器26的通道部分28周圈形成的锐利尖头30处。锐利尖头30是以一个角度截成的圆筒形节段形式,因而在锐利尖头的外表面和燕管夹持器26的内表面之间形成一个圆筒形间隔32,以在芯管10依箭头34的方向被推入至图3B中所示的位置时接纳圆筒体12的一部分。
芯管夹持器26的内表面和芯管10设计成当芯管10处于图3B所示的位置时,芯管10就被形成芯管夹持器26的圆筒壁夹持在其位置上。当将芯管朝着箭头34的方向推动时,尖头30就刺穿芯管10里端的铝箔层18而使其敞开在烟嘴22的通道28之前。
为了使空气能流过芯管10内并经过或穿过存贮体16,在芯管10外端的尼古丁不透层18也必须刺穿,这可以用刀子之类的任何尖锐的东西来完成。然而,提供一种容易使用的尖锐物件的方式就是提供一个如图4所示那种型式的分配器36,它是由塑料模制而成并包括有多个用于容纳芯管10(未显示)的隔室38和一个用于放置烟嘴22的凹槽40。所有这些部件都可以缩包在一张透明塑料薄片内,用作销售包装。
为了提供一个在手边的尖锐物件用以刺穿芯管10外端的箔层18,放置烟嘴22的凹槽40的一端可以设置一个锋利的尖头42。这样,在芯管10插入烟嘴22的端头并被推至图3B所示的位置后,其外端只要用力顶压在锋利的尖头42上就能将其刺穿,如图4所示。这样,通道28就能通过芯管10的通道14与外界相通,因而吸用者只要吮吸烟嘴22的口衔部分24就能获得所述的尼古丁气化物。
另一种使用锋利的尖头来刺穿其中一个或两个箔端的方式是在箔上配置一个能用手抓住的部分(未显示),以便使使用者能将箔层18从芯管10上撕去。
本发明的另一个实施例示于图5和6中,其中一个其构形与先前所述相同的芯管10与一个芯管刺穿器/盖44结合使用。如图5 A所示,刺穿器/盖44被插套在芯管10的外端上,此组合件随之插入到烟嘴2 2的芯管夹持器26的外端,与图2和3中所示者相似。
芯管刺穿器/盖由一个圆筒体46形成,该圆筒形成一条通道48,通道的外端由一个圆筒形锋利尖头50限定,该锋利尖头的构形与芯管夹持器26中的锋利尖头30是一样的。在锋利尖头50的外表面与圆筒体46的内表面之间形成有一环形间隔52,用于接纳芯管10的圆筒体12。
在将刺穿器/盖44放置在芯管10的外端上后,将它沿着箭头54(图5B)的方向朝着图5中所示的位置推动,从而使锋利尖头50刺穿位于芯管10外端的箔层18。这样,烟嘴22的通道28和芯管10的通道14互相连通并与外界相通,因而吸用者能够吮吸烟嘴而获得如前所述的尼古丁气化物。
图5和图6中所示的本发明实施例可以用图7中所示的方式予以包装,其中一个用塑料模制的盘56包括多个隔室58,用以盛装如图5A中所示的处于未经刺穿的位置的芯管和芯管刺穿器/盖44。还可以设置一个隔室60以盛装烟嘴22,所有的一切元件均可用透明塑料(未显示)缩包起来的方式进行包装。
使用了上述的吸入器,一个与烟嘴结合使用的装有尼古丁的芯管就可以上市场销售,即便经过一个不一定是很短的贮存期限也不致丧失其效能。通过在芯管上装配了一个尼古丁不能透过的阻挡体,尼古丁就不会从芯管中逸出,而开始时装入的剂量可以在该产品的整个使用寿命期间始络维持不变。还有,通过将尼古丁存贮体维持和存放在一个无氧的环境中,尼古丁就不会由于与氧相互作用而变质,尼古丁剂量的有效程度也可维持不变。
上面的描述,其性质并不是限制性的,本发明还应当包括除了上面具体描述的之外的、在后附的权利要求的精神和范围之内的一切改进和变异。

Claims (14)

1.一种用于尼古丁吸入器的芯管,它包括:
a)一个芯管外壳(10);
b)在所述芯管外壳(10)内的通道(14);
c)在所述通道(14)内的一个用于盛装定量的尼古丁的尼古丁存贮体(16),其形式能容许尼古丁气化物释放在绕经或通过存贮体(16)的流体流中;
d)所述通道(14)包括有至少两个与外壳的外面相通的开口以容许流体流穿过通道(14);和
e)所述尼古丁存贮体(16)是由一个尼古丁不能透过的阻挡体(18)将其与外界隔绝,该阻挡体包括多个用于将通道在存贮体(16)的两侧封住的通道阻挡体部分,至少所述通道阻挡体部分是可以被穿透以使通道(14)与外界相通;
f)其中在所述通道阻挡体部分里面的所述通道(14)的部分内是无氧的。
2.如权利要求1的芯管,其特征在于:其中芯管外壳(10)是一个细长件,通道(14)由此细长件的内表面形成,而通道的开口位于细长件的两相对端。
3.如权利要求1或2的芯管,其特征在于:其中的细长件是圆筒形状的。
4.如权利要求1至3中任一项的芯管,其特征在于:其中的尼古丁存贮体(16)包括一个装有尼古丁游离碱的海绵体聚合物塞头。

9.如权利要求1至8中任一项的芯管,其特征在于:其中尼古丁不能透过的阻挡体(18)包括以一层铝箔包覆所述芯管外壳(10)。
10.如权利要求9的芯管,其特征在于:其中的铝箔包括至少在其一边涂有丙烯腈和丙烯酸甲酯的共聚物的涂层。
11.如前述权利要求中任一项的芯管,其特征在于:其中在所述通道阻挡体部分里面的所述通道(14)的部分内充以惰性气体,例如氮。
12.一种尼古丁吸入器,该吸入器包括一个芯管和一个烟嘴;
该芯管包括:
a)一个芯管外壳(10);
b)在所述芯管外壳(10)内的通道(14);
c)在所述通道(14)内的一个用于盛装定量的尼古丁的尼古丁存贮体(16),其形式能容许尼古丁气化物释放在绕经或通过存贮体(16)的流体流中;
d)所述通道(14)包括有至少两个与外壳的外面相通的开口以容许流体流穿过通道(14);和
e)所述尼古丁存贮体(16)是由一个尼古丁不能透过的阻挡体(18)将其与外界隔绝,该阻挡体包括多个用于将通道在存贮体(16)的两侧封住的通道阻挡体部分,至少所述通道阻挡体部分是可以被穿透以使通道(14)与外界相通;

将已装料的存贮体(16)在无氧的状况下装入所述芯管外壳(10)中;和
用尼古丁不能透过的阻挡体(18)将已装料的存贮体(16)密封在通道(14)内,阻挡体(18)包括密封住存贮体两侧的通道的通道阻挡体部分,至少所述通道阻挡体部分是可以被刺穿的以便使所述通道通向外界。
14.如权利要求13的方法,其特征在于:其中装料的步骤还包括下述步骤:
将尼古丁存贮体(16)置入真空室内;在所述室内造成一个负压;向所述负压室内加进一种尼古丁,薄荷醇,乙醇溶液;和在无氧的环境中将所述已装料的存贮体(16)从所述室中取出。
